In order to explore the risk factors of infertility caused by endometriosis (EMT) and their correlation with vascular endothelial growth factor (VEGF), TNF-α, IL-6, IL-10, and IL-17, endometriosis sufferers admitted to our hospital from January 2021 to May 2022 are selected to conduct the examination. According to the pregnancy of patients, patients were included in the simple EMT set and EMT combined infertility set, with 50 cases in each group. The degree of dysmenorrhea is evaluated by the VAS score, and Luminex liquid protein is used to analyze the standards of the tumor necrosis factor (TNF-A), interleukin (IL)-10, IL-6, IL-17, and VEGF. Logistic multifactor regression decomposition is applied to analyze the risk factors of infertility in EMT sufferers. Besides, the standards of VEGF, TNF-α, IL-6, IL-10, and IL-17 in sufferers with different periods/agony degrees are evaluated, and the correlation of different periods/agony degrees with VEGF, TNF-α, IL-6, IL-10, and IL-17 is analyzed. The results show that the different R-AFS periods are notoriously positively correlated with VEGF, TNF-α, IL-6, IL-10, and IL-17 (all P < 0.05 ), and the VAS score is notoriously positively correlated with the abovementioned factors.

IntroductionEndometriosis (EM) is an aggressive, pleomorphic, and common gynecological disease. Its clinical presentation includes abnormal menstruation, dysmenorrhea, and infertility, which seriously affect the patient's quality of life. However, the pathogenesis underlying EM and associated regulatory genes are unknown.MethodsTelomere-related genes (TRGs) were uploaded from TelNet. RNA-sequencing (RNA-seq) data of EM patients were obtained from three datasets (GSE5108, GSE23339, and GSE25628) in the GEO database, and a random forest approach was used to identify telomere signature genes and build nomogram prediction models. Gene Ontology, Kyoto Encyclopedia of Genes and Genomes, and Gene Set Enrichment Analysis were used to identify the pathways involved in the action of the signature genes. Finally, the CAMP database was used to screen drugs for potential use in EM treatment.ResultsFifteen total genes were screened as EM–telomere differentially expressed genes. Further screening by machine learning obtained six genes as characteristic predictive of EM. Immuno-infiltration analysis of the telomeric genes showed that expressions including macrophages and natural killer cells were significantly higher in cluster A. Further enrichment analysis showed that the differential genes were mainly enriched in biological pathways like cell cycle and extracellular matrix. Finally, the Connective Map database was used to screen 11 potential drugs for EM treatment.DiscussionTRGs play a crucial role in EM development, and are associated with immune infiltration and act on multiple pathways, including the cell cycle. Telomere signature genes can be valuable predictive markers for EM.

Introdução: Hoje, sabe-se que a endometriose é uma doença ginecológica crônica, de caráter inflamatório, marcada pela presença de tecido endometrial fora do útero (ectópico), dependente de estrogênio. A endometriose pode ser classificada em superficial, ovariana e profunda, além da extrapelvica e é marcada principalmente por dispareunia, dismenorreia, disuria, disquezia e infertilidade (um desafio para 30 a 50% dessas mulheres). Objetivo: Identificar os mecanismos etiopatogênicos que poderiam justificar a prevalência da infertilidade em mulheres com endometriose. Materiais e Métodos: Trata-se de uma revisão integrativa de literatura acerca das novas evidências que justificam a infertilidade em mulheres com endometriose. Utilizou-se a estratégia PICO para a elaboração da pergunta norteadora. Ademais, realizou-se o cruzamento dos descritores “endometriose”, “infertilidade”, “patogênese” e “etiologia”, nas bases de dados National Library of Medicine (PubMed MEDLINE), Scientific Eletronic Library Online (SCIELO), Google Scholar e Biblioteca Virtual de Saúde (BVS). Resultados e Discussão: O ambiente proporcionado pela endometriose promove diversas alterações, entre elas: o alto caráter inflamatório, o desbalanço de citocinas, o estresse oxidativo e alteração do fluido folicular e do peritoneal. O sistema imune parece estar prejudicado, com alteração da função da maioria das células imunes. Desbalanço hormonal, fatores genéticos e epigenéticos também são fundamentais. Conclusão: A associação entre a infertilidade e a endometriose parece prejudicar a concepção devido a alteração do fluido folicular e da foliculogênese, a redução da qualidade dos oocitos, a alteração da janela de implantação, a alterações espermáticas e a disfunção de células ciliares.
