Endogenous retroviruses drive KRAB zinc-finger protein family expression for tumor suppression

Abstract: Gene expression aberration is a hallmark of cancers, but the mechanisms underlying such aberrations remain unclear. Human endogenous retroviruses (HERVs) are genomic repetitive elements that potentially function as enhancers. Since numerous HERVs are epigenetically activated in tumors, their activation could cause global gene expression aberrations in tumors. Here, we show that HERV activation in tumors leads to the up-regulation of hundreds of transcriptional suppressors, namely, Krüppel-associated box domain… Show more

“…TEeRS can act as enhancers, repressors, promoters, terminators, insulators or via post-transcriptional mechanisms (Garcia-Perez et al 2016;Chuong et al 2017). Although these co-opted TE functions are key to human biology, their deregulation can also contribute to pathologies such as cancer and neurodegenerative diseases (Li et al 2015;Chuong et al 2016;Attig et al 2019;Jang et al 2019;Ito et al 2020;Jönsson et al 2020).…”

“…It seems that cancer and immune diseases are not accompanied by transcriptional activation of all HERVs, but rather the upregulation of specific HERV loci in different diseases was observed. Other studies described a more global activation of HERV especially in solid tumor samples ( 25 ). Nevertheless, different tumor entities could be classified according to their HERV expression ( 25 ) suggesting entity specific expression pattern.…”

“…Other studies described a more global activation of HERV especially in solid tumor samples ( 25 ). Nevertheless, different tumor entities could be classified according to their HERV expression ( 25 ) suggesting entity specific expression pattern. Due to the large group of HERVK it cannot be excluded that the differential expression of single loci is not detected by our evaluation.…”
