Endogenous Retrovirus 3 – History, Physiology, and Pathology

Abstract: Endogenous viral elements (EVE) seem to be present in all eukaryotic genomes. The composition of EVE varies between different species. The endogenous retrovirus 3 (ERV3) is one of these elements that is present only in humans and other Catarrhini. Conservation of ERV3 in most of the investigated Catarrhini and the expression pattern in normal tissues suggest a putative physiological role of ERV3. On the other hand, ERV3 has been implicated in the pathogenesis of auto-immunity and cancer. In the present review … Show more

“…Inhibition of methyltransferases is able to activate ERV expression in cancer cells with subsequent induction of anti-viral pathways [29]. This so-called “viral mimicry” can be considered as a natural defense mechanism against transformed or virus-infected cells [30]. A binary switch model has been proposed for histone regulation [31].…”

“…The role of ERV3-1 in cancer appears to be different in several tumor entities. The overexpression of ERV3-1 is associated with several tumor entities like prostate, lung, liver and colorectal cancer ( 8 , 64 , 70 ). However, in the myelogenous leukemia cell line U-937 ERV3 was found to be upregulated during monocyte differentiation ( 71 ) because of demethylation of the ERV3 locus ( 72 ).…”

“…HERV-R transcripts have broad expression in normal tissue and due to its overexpression in the first trimester of pregnancy an immunosuppressive function in the mother-fetus interaction was suggested (54,55). ERV3-1 is the only copy of approximately 40 ERV3-like elements in the human genome that has a complete ORF for its ENV (56). Interestingly, the expression of this full-length ENV is associated with several tumor entities including colorectal (57), ovarian (58) and endometrial carcinoma.…”
