Endocrine and Hemodynamic Effects of Stress Versus Systemic CRF in Alcoholics during Early and Medium Term Abstinence

Abstract: In alcoholics, disturbances of the autonomic nervous system as well as of the hypothalamic-pituitary-adrenal axis (HPA) are known. However, these two systems have never been analyzed, under stimulated conditions, in parallel in the same patients. Moreover, studies using intravenous (i.v.) corticotropin releasing factor (CRF) to assess neuroendocrine function bypass the hypothalamic component of the HPA axis. Therefore, i.v. human (h) CRF (pituitary stimulation/exogenous CRF) and a multifaceted stress test (hyp… Show more

“…Blood was drawn into EDTA-coated monovettes (ethylenediaminetetraacetic acid; Sarstedt, Numbrecht, Germany), and immediately centrifuged for 10 min at 2,000g; plasma was stored at -80°C until analysis. Plasma norepinephrine was determined by means of HPLC and electrochemical detection after liquid-liquid extraction (Smedes et al, 1982;Ehrenreich et al, 1997). The limit of detection was 10 pg/ml.…”

“…Catecholamine plasma levels were measured by HPLC (Ehrenreich et al 1997, Smedes et al 1982. The intraand interassay variability for noradrenaline were 6·2 and 8·0% respectively, and 4·0 and 5·1% respectively for adrenaline.…”

“…The samples were stabilized with EDTA (5 mM) and stored at 7208C. Liquid-liquid-extraction of the samples (Smedes et al, 1982) was performed as previously described (Ehrenreich et al, 1997) using an extraction kit (Variokit, Biometra, GoÈ ttingen, Germany). In brief, to 0.5 ml of the sample, 20 ml (500 pg) dihydroxybenzoic acid (DHBA internal standard) and 150 ml of NH 4 OH/NH 4 Cl bu er, containing diphenylborate-ethanolamine and EDTA were added.…”
