Aim of the study was to develop optimised drug-in-adhesive (DIA) transdermal patch of duloxetine HCl. It is known that acrylic polymers having different functional groups play significant role in enhancing drug permeation. Among various permeation enhancers (PEs), Transcutol P exhibited most enhanced permeation (ER≈1.99) in terms of flux and Q24 compared to control group having no PE. Hence, a transdermal DIA patch having DURO-TAK 87-2287 as DIA polymer and TP as PE loaded with 40% DLX previously complexed with MeβCD and duly characterised (FTIR, DSC and SEM) was developed for in vivo study. Mean of maximum plasma concentration (Cmax) and area under time-concentration curve (AUC0-72) in Wistar rats (n=6) for transdermal patch (10 mg/kg) was found to be 70.31±11.2 ng/ml and 2997.29±387.4 ng/ml*h respectively and these values were considerably higher than oral dose of DLX (20 mg/kg and 10 mg/kg). Albeit, T1/2 was higher in case of transdermal delivery but this was due to sustained behaviour of delivery system. These findings highlight the significance of both inclusion complexation and transdermal delivery of DLX using DIA patch for efficient drug absorption and thus reducing the dose of drug and related side effects.

β-Cyclodextrin Inclusion Complex Containing Litsea cubeba Essential Oil: Preparation, Optimization, Physicochemical, and Antifungal Characterization

Litsea cubeba essential oil (LCEO), as naturally plant-derived products, possess good antimicrobial activities against many pathogens, but their high volatility and poor water solubility limit greatly the application in food industry. In this research, inclusion complex based on β-cyclodextrin (β-CD) and LCEO, was prepared by saturated aqueous solution method. An optimum condition using the response surface methodology (RSM) based on Box–Behnken design (BBD) was obtained with the inclusion time of 2 h and β-CD/LCEO ratio of 4.2 at 44 °C. Under the condition, the greatest yield of 71.71% with entrapment efficiency of 33.60% and loading capacity of 9.07% was achieved. In addition, the structure and characteristic of LCEO/β-CD inclusion complex (LCEO/βCD-IC) were investigated using scanning electron microscopy (SEM), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FT-IR), which indicated that LCEO/βCD-IC was successfully formed. The particle size of LCEO/βCD-IC was determined to be 17.852 μm. Thermal properties of LCEO/βCD-IC evaluated by thermogravimetric-differential scanning calorimetry (TG-DTA) illustrated better thermal stability of the aimed product compared with the physical mixture. Furthermore, the tests of antifungal activity showed that LCEO/βCD-IC was able to control the growth of Penicillium italicum, Penicillium digitatum, and Geotrichum citri-aurantii isolated from postharvest citrus. Our present study confirmed that LCEO/βCD-IC might be further applied as an alternative to chemical fungicides for protecting citrus fruit from postharvest disease.

The present study discussed the optimization of the ultrasonic‐assisted extraction of polysaccharides from daylily polysaccharides (DPs). The extracted crude polysaccharides were further separated and purified, and the antioxidant activities including 1,1‐diphenyl‐2‐111 picrylhydrazyl (DPPH) radical scavenging, 2,2'‐azinobis‐(3‐ethylbenzthiazoline‐6‐sulphonate) (ABTS) radical scavenging, hydroxyl radical scavenging, and ferric‐reducing antioxidant power (FRAP) activities of the obtained fractions were also evaluated. The results showed that the optimal ultrasonic‐assisted extraction parameters with DPs yield of 15.25 ± 1.13% were water to powder ratio of 25 ml/g, extraction power of 694 W, extraction temperature of 71°C, extraction time of 38 min, and three times extraction. By DEAE Sepharose Fast Flow column, four water‐soluble polysaccharide fractions (DP‐1, DP‐2, DP‐3, and DP‐4) were successfully obtained. Monosaccharide component analysis showed that the four obtained fractions were all hetero‐polysaccharides that mainly contained rhamnose, arabinose, fructose, galactose, glucose, galacturonic acid, and glucuronic acid in different molar ratios. All the four DP fractions did show obvious antioxidant activities in vitro, and the DP‐3 component had relatively high ABTS free radical scavenging activity. Overall, our research showed that DPs could provide cheap raw materials for the development of natural antioxidants in medicines, functional foods, and even cosmetics. Practical Application This article deals with the optimization of the ultrasonic‐assisted extraction of polysaccharides from daylily and its antioxidant activities. The results showed that the optimal ultrasonic‐assisted extraction yield of DPs was 15.25 ± 1.13%. By DEAE Sepharose Fast Flow column, four water‐soluble polysaccharide fractions were successfully obtained, and all the four DP fractions did show obvious antioxidant activities in vitro. Daylily polysaccharides could provide cheap raw materials for the development of natural antioxidants in medicines, functional foods, and even cosmetics.
