Encapsulation,In VitroCharacterization, andIn VivoBiocompatibility of Sertoli Cells in Alginate-Based Microcapsules

Abstract: A method for microencapsulation of isolated neonatal porcine Sertoli cells is described. Using a conventional alginate-poli-L-ornithine encapsulation procedure, which has been used in our laboratory for almost two decades to envelop pancreatic islets, we observed significant loss of Sertoli cell viability, possibly due to excessive Ca(2+) ion exposure. Replacing calcium with barium, or shortening the incubation period in the presence of Ca ions, we obtained barium or calcium alginate gel microbeads that did no… Show more

“…Ions belonging to the second group of the Mendeleev's periodic table share the ability to ligate alginate. Barium alginate microcapsules are mechanically stable and easy to obtain, with a procedure that is substantially independent of the type of cells to be encapsulated [101,110].…”

“…SCs were isolated from the testes of neonatal pre-pubertal ''Large-White" pigs, aged 7-15 days, according to previously reported methods and as modified in our laboratory [12]. Briefly, decapsulated testicular tissue was minced and then digested with trypsin (2 mg ml À1 ) and DNAse I (Sigma) to obtain a clean fraction of seminiferous tubules devoid of other testicular cells such as Leydig cells and peritubular cells.…”

“…After washing the Tris off, the SCs were cultured under the same conditions, in 75 cm 2 cell culture flasks. After isolation, purified SC monolayers were identified by anti-Mullerian inhibiting substance (MIS) immunostaining, according to previous reports [12]. Cell viability was assessed, before and after encapsulation, by double staining with PI and Calcein-AM according to the manufacturer's instructions.…”

“…Highly monodisperse hydrogel microbeads have many important applications in pharmacy (Nortier et al 1995), food coatings (Sanguansri et al 2006) and biomedical industries such as cell encapsulation (Jen et al 1996;Luca et al 2007) and drug delivery systems (Graham and Mcneill 1984). In cell encapsulation for tissue engineering and cell-based beads array systems, micro-sized monodisperse hydrogel beads are in demand since they facilitate handling and allow rapid exchange of nutrients into and waste out of these beads.…”
