Enantioselective Total Synthesis of Sacubitril

Abstract: An efficient enantioselective approach of Sacubitril 1 (a NEP inhibitor) which is used as a combination drug with Valsartan to treat heart failure, is described. The synthesis features the Evan's asymmetric alkylation, Sharpless asymmetric diydroxylation (AD) and Heck coupling reactions as key steps.
