Enantiopure Chiral Poly(glycerol methacrylate) Self‐Assembled Monolayers Knock Down Protein Adsorption and Cell Adhesion

Abstract: Chirality plays a fundamental role not only in biological systems, but also in synthetic materials intended for bio-applications. Self-assembled monolayers (SAMs) are prepared on gold surfaces through a "grafting to" method from racemic or enantiopure chiral poly(glycerol methacrylate)s (PGMA(rac), PGMA(R), and PGMA(S)), having a thiol endgroup. Such SAMs constitute a chemically and structurally well-defined model substrate for studying protein adsorption and cell adhesion as a function of the polymer chiralit… Show more

“…To determine how FoxO3 translocation varies across cell lines, we selected, from a panel of widely studied breast cancer cells, seven lines that include HER2 AMP , hormone-receptor positive, and triple negative subtypes (the ICBP43 set (Li et al, 2013)); 184A1 and MCF10A cells were included as examples of normal mammary epithelial controls. Mutations in mitogenic signaling networks are common in breast cancer, and multiple cell lines in our collection carry oncogenic mutations in HRAS, KRAS, BRAF and PIK3CA.…”

“…D-sugars and Lamino acids). Although initial observations of surface chiralityrelated protein adsorption [15][16][17] and cellular adhesion 18,19 have been reported, molecular level understanding of how material surface chirality can inuence protein assembly (e.g. the dynamics of adsorption, kinetics of conformational transition and bril formation), and its consequent chirality-dependent biological effects remains unexplored.…”

“…Many chiral superstructures can be self-assembled from chiral or achiral molecules, and these chiral superstructures may be used in various fields as templates for helical crystallization, molecular recognition, catalysis and so on 9 10 11 12 . Recently, pioneering works have been conducted to reveal the cell behaviors such as cell adhesion 13 and differentiation 14 , and protein adsorption 15 16 ( e.g. amount and affinity) on flat substrates anchored with different chiral molecules.…”
