Emerging Roles of Meningeal Lymphatic Vessels in Alzheimer’s Disease

Abstract: Meningeal lymphatic vessels (mLVs), the functional lymphatic system present in the meninges, are the key drainage route responsible for the clearance of molecules, immune cells, and cellular debris from the cerebrospinal fluid and interstitial fluid into deep cervical lymph nodes. Aging and ApoE4, the two most important risk factors for Alzheimer’s disease (AD), induce mLV dysfunction, decrease cerebrospinal fluid influx and outflux, and exacerbate amyloid pathology and cognitive dysfunction. Dysfunction of mL… Show more

“…How PBM can improve cognitive function remains poorly understood. Emerging evidence indicates the important roles of BDS in the resistant to neurocognitive disorders [ 8 , 9 , 14 , 63 68 ]. Indeed, dysfunction of MLVs results in less drainage of brain fluids to the cervical lymph nodes [ 8 , 11 , 12 , 19 , 28 , 41 , 42 ].…”

“…Such disruption also results in cognitive impairment and behavioral alterations due to dysfunctions of the brain’s waste removal system leading to the accumulation of toxins and metabolites in the CNS 75 . The dysfunction of the MLVs might provide an important contribution to age-related cognitive decline and neuro-degenerative disease 76 , 77 . The MLVs also affect microglia responses 75 .…”

“…In MLVs specifically, such age-related dysfunctions reduce influx and outflux of CSF. It triggers the accumulation of metabolic products, accelerates neuroinflammation, induces the release of pro-inflammatory cytokines in the brain, and consequently aggravates cognitive dysfunction (Guo et al 2023 ).…”
