Emerging roles of HOTAIR in human cancer

Abstract: Long noncoding RNA HOX antisense intergenic RNA (HOTAIR) is overexpressed in many types of cancers, and substantial evidence has suggested a link between cancers and HOTAIR. In the present study, we reviewed the structure and the corresponding biologic function of HOTAIR to clarify its molecular mechanism in cancer progression. HOTAIR promotes proliferation, invasion, and migration, and inhibits apoptosis in cancer cells. HOTAIR also participates in the pathogenesis and progression of cancer by regulating infl… Show more

“…HOX transcript antisense intergenic RNA (HOTAIR) is a 2158-bp LncRNA transcribed from the antisense strand of the homeobox C gene on chromosome 12 [4]. HOTAIR acts in trans on the HOX D locus of chromosome 2 through chromatin-remodeling enzymes [5]. It is a novel prognostic biomarker, especially for the potential metastatic prediction [6].…”

“…Few studies have examined the effects of steroids, including P4, on lncRNA expression in cancer development. lncRNAs such as HOX transcript antisense intergenic RNA (HOTAIR) 35 and T‐cell leukemia/lymphoma 6 (TCL6) 36 were reported to be associated with the PR status of breast cancer, and may predict disease prognosis. lncRNAs such as metastasis‐associated lung adenocarcinoma transcript 1 (MALAT1) 37 and nuclear enriched abundant transcript 1 (NEAT1) 38 have been shown to regulate the cell cycle and influence the progression of endometrial cancer.…”

“…HOTAIR (HOX antisense intergenic RNA); this lncRNA acts as a scaffold that recruits chromatin modifying complexes PRC2 (polycomb repressive complex 2) to the HOXD locus, coordinates H3K27 methylation and H3K4me2 demethylation of target genes, and contributes to maintaining a repressive chromatin state [ 27 ], which has been implicated in the pathogenesis of several kinds of cancers, including breast cancer, lung cancer, and colon cancer [ 28 ]. On the other hand, HOTTIP, which is transcribed from a locus upstream of HOXA genes, recruits the MLL-1/WDR5 complex to the 5’ region of HOXA genes, mediates H3K4me3 modification and activates transcription of the HOXA locus [ 29 ].…”
