Emerging role of SIRT3 in endothelial metabolism, angiogenesis, and cardiovascular disease

Abstract: Sirtuin 3 (SIRT3) a mitochondrial enzyme that plays an important role in energy homeostasis, cardiac remodeling, and heart failure (HF). The expression of SIRT3 declines with advanced age, cardiovascular, and metabolic diseases. Accumulating evidence suggests that SIRT3 plays a critical role in protecting the heart from cardiac hypertrophy, cardiac dysfunction associated with HF, and in the protection of cardiac cells from stress-mediated cell death. Clinical studies have demonstrated that HF with preserved ej… Show more

“…Sirtuins are important regulatory elements of energy metabolism and antioxidative defense, using post-translational modifications [32,52,73,74]. Sirtuins were recently shown to regulate vascular and especially endothelial functions, under physiological conditions [1][2][3][4]. Dysregulation of sirtuins was observed under pathophysiological conditions like cardiovascular disease [20][21][22][23][24] and vascular aging, including atherosclerosis [1,[25][26][27][28].…”

“…Rodents and humans demonstrate the critical role of SIRT3 in metabolic diseases, highlighting the decrease in SIRT3 activity in several tissues such as the liver, the adipose tissue and the muscle [174,175]. Mice deficient for SIRT3 on an HFD also reveal accelerated hallmarks of MetS [176].…”

“…SIRT3 has been shown to blockade cardiac hypertrophy and attenuate aging and oxidative stress-mediated cell death in cardiomyocytes via Foxo3a and Ku70 (24). In addition, SIRT3 deficiency impairs mitochondrial function and cardiac function by hyperacetylation of energy metabolic proteins and myocardial energy depletion (16, 17).…”
