Emerging Hallmarks of Metabolic Reprogramming in Prostate Cancer

Abstract: Prostate cancer (PCa) is the most common male malignancy and the fifth leading cause of cancer death in men worldwide. Prostate cancer cells are characterized by a hybrid glycolytic/oxidative phosphorylation phenotype determined by androgen receptor signaling. An increased lipogenesis and cholesterogenesis have been described in PCa cells. Many studies have shown that enzymes involved in these pathways are overexpressed in PCa. Glutamine becomes an essential amino acid for PCa cells, and its metabolism is thou… Show more

“…In recent years, the introduction of high-throughput technologies has provided significant insights into the understanding of normal and pathological processes, as well as the identification of potential targets for the development of drugs to treat diseases such as cancer [55][56][57][58][59][60][61]. In particular, the analysis of low-molecular-weight metabolites (the metabolome) allows for the global assessment of a cellular state in normal and pathological conditions.…”

“…Fibroblasts are the major components of the stroma of PC (Pederzoli et al 2023 ). Fibroblasts are necessary for cancer progression, cancer metastasis, and being androgen-independent (Lasorsa et al 2023 ). A recent study was conducted on three prostate cancer cell lines (PC-3 and DU-145, and LNCaP) and healthy prostate cells as control (PNT-1) (Pietrovito et al 2020 ).…”

“…ARG1-expressing cells also include M2-TAMs and T regs [ 47 ]. Reduced levels of these amino acids impair T cell proliferation [ 48 , 49 ]. In addition, regulatory T cells (Treg) may dampen T cell responses at tumor sites and lymph nodes.…”
