Emergence of dynamic cooperativity in the stochastic kinetics of fluctuating enzymes

Abstract: Dynamic cooperativity in monomeric enzymes is characterized in terms of a non-Michaelis-Menten kinetic behaviour. The latter is believed to be associated with mechanisms that include multiple reaction pathways due to enzymatic conformational fluctuations. Recent advances in single-molecule fluorescence spectroscopy have provided new fundamental insights on the possible mechanisms underlying reactions catalyzed by fluctuating enzymes. Here, we present a bottom-up approach to understand enzyme turnover kinetics … Show more

“…Whereas competing parallel and off‐parallel corridors led to a broad and highly stretched distribution function for the network. We here emphasize once again that dynamic disorder arises in this case even in the absence of a distinct inhibitor as discussed in a few recent articles 10,20,21 …”

“…However, one can extract valuable information on the kinetic mechanisms of a single enzyme from the inverse of the randomness n=1/r and associate it to the number of rate‐limiting steps of the kinetic cycle, including identification of off‐pathways within the network if any. 10,19,20 So r=1/n is the minimum amount of randomness that can be summarized. Hence, r>1 or r<1 and r=1 in the perspective of statistical kinetics brings insight into the emergent features of the dynamics at the stochastic level.…”
