This study evaluated the vulnerability of early life stages of native oysters (Crassostrea gigas) from the Arcachon Bay (SW, France) to pollutants at risk in the lagoon in particular copper and S-metolachlor. Developmental abnormalities in wild and cultivated oyster D-larvae were investigated during 2 breeding-seasons (2013 and 2014) at different sampling sites and dates. In addition, copper, and metolachlor concentrations were determined both in seawater and in mature oysters. Bioaccumulation of Cu was observed at higher levels in wild than in farmed specimens. Metolachlor was accumulated at much lower levels. After 24 h exposure, significant increases of the percentage of abnormal D-larvae were observed when exposed at 1 µg L −1 of copper or 10 ng L −1 of metolachlor in comparison with the controls whatever the date, the site, and rearing conditions of the genitors. The current study demonstrates that environmental concentrations of copper and metolachlor can induce a significant increase of developmental abnormalities in farmed and wild populations of oysters. However, no significant differences of sensitivity were observed according to temporal, spatial parameters, and lifestyle of genitors. In addition, oyster larvae obtained from the hatchery displayed the same sensitivity to pollutants as larvae from the field, demonstrating their suitability for toxicity assays and water quality monitoring.

N-Nitrosodimethylamine (NDMA) was shown to have genotoxic properties in acute and subacute studies on laboratory mice. The organ-specificity of the genotoxic effect of NDMA was revealed using the Comet assay. The most sensitive organs to the action of NDMA were kidneys and liver. DNA damage in liver cells of NDMA-treated animals at doses of 4.0 and 8.0 mg/kg, increased compared to control in 6.9 and 12.5 (р < 0.001), and in kidney cells – in 8.1 and 14.2 times (р < 0.001), respectively. NDMA also showed genotoxic activity in the reproductive cells of experimental animals, causing structural disorders of synaptonemal complexes in spermatocyte. In NDMA-treated animals at a dose of 2.0 mg/kg in acute and subacute studies, the level of spermatocytes with damaged synaptonemal complexes increased statistically significantly compared to control in 6.0 and 7.0 (р < 0.05) times, respectively.

Copper (Cu) is an essential trace metal, but may also be toxic to aquatic organisms. Although many studies have investigated the cytotoxicity of Cu, little is known about the in vivo genotoxic potential of Cu in marine invertebrates. We investigated the genotoxicity of Cu in 2 pelagic calanoid copepods, Acartia tonsa and Temora longicornis, and the intertidal harpacticoid copepod Tigriopus brevicornis by exposing them for 6 and 72 h to waterborne Cu (0, 6, and 60 µg Cu/L). A subsequent 24‐h period in filtered seawater was used to investigate delayed effects or recovery. Genotoxicity was evaluated as DNA strand breaks in individual copepods using the comet assay. Copper did not increase DNA strand breaks in any of the species at any concentration or time point. The treatment did, however, cause 100% mortality in A. tonsa following exposure to 60 µg Cu/L. Acartia tonsa and T. longicornis were more susceptible to Cu‐induced mortality than the benthic harpacticoid T. brevicornis, which appeared to be unaffected by the treatments. The results show major differences in Cu susceptibility among the 3 copepods and also that acute toxicity of Cu to A. tonsa is not directly associated with genotoxicity. We also show that the comet assay can be used to quantify genotoxicity in individual copepods. Environ Toxicol Chem 2019;38:2224–2232. © 2019 SETAC.
