Mammillaria lasiacantha is a small cactus that inhabits at Sierra of Juárez, Chihuahua, Mexico. Their populations have been affected by habitat destruction, illegal extraction, and uncontrolled tourism in the region that steps on the seedlings; therefore, knowing its reproductive biology is necessary for its conservation. Our main questions were: What is the floral morphology and anatomy like? What type of sexual and mating system do M. lasiacantha present? Do the seeds produced by the crosses germinate, how and in what percentage? Sixty individuals from a population of 120 plants were studied at Sierra of Juárez, Juárez Chihuahua from January to August 2021. The study of the floral morpho-anatomy was carried out. Four treatments were applied: natural selfing, forced selfing, geitonogamy and forced outcrossing plus a control. Pollinated flowers, fruits, number of seeds and germination percentage were recorded. Flowers have naked pericarp, funnel-type nectary, nectarostomata, closed style and collateral vascular bundles. The anatomical characteristics of M. lasiacantha agree with those reported for other species of Mammillaria. The flowers of M. lasiacantha present herkogamy, with an obligate outcrossing system. The seeds produced from the outcrossing and control group germinated in 32 and 76%, respectively. The herkogamy and strict outcrossing system of M. lasiacantha should be confirmed in other populations. The seeds are positive photoblastic, without dormancy, but the study of the seed bank and seedling survival in the field would be pertinent. The ex-situ conservation of M. lasiacantha in the site will be feasible, since species high percentage of germination may contribute to produce plants for restoration programs.

Seeds of 13 Pachycereus species and two Stenocereus species that have been suggested as closely related were examined with the scanning electron microscope. Quantitative features were evaluated using multivariate analysis in order to identify characters that distinguish them. Several species groups were recognized on the basis of 16 qualitative characters. All species studied are keeled. Stenocereus aragonii and S. eichlamii share with most Pachycereus species large size, glossy appearance, and a flat relief on periclinal cells in the lateral region. Pachycereus gatesii and P. schottii are unique in having the smallest seeds and a deeply impressed hilum-micropylar region. P. hollianus does not exhibit micro-relief on periclinal walls in the lateral region, and P. fulviceps has no expanded testa border. Multivariate analysis showed that four characters, length, breadth, hilum-micropylar region length, and angle, made the greatest contribution to distinguishing among species groups. More than 80% of P. fulviceps, P. hollianus, P. tepamo, P. weberi, and S. eichlamii seeds could be classified correctly using four seed features and the percentage was even higher using just two or three features for P. gatesii, P. grandis, P. militaris, P. pringlei, and P. schottii. Testa appearance, testa cell-pattern, and position relative to the rim of the hilum-micropylar region were found to be potentially informative and should be combined with other sources of data in future phylogenetic analyses.

The structural details of the gynoecium are key to understanding the reproductive systems and successful diversification of flowering plants. However, the gynoecium morpho-anatomy in South American species of Cactaceae that evolved in the Andean region remains largely unknown. Here we selected 18 species, most of them of evolutionarily related genera of Cactaceae tribe Trichocereeae, to conduct a detailed comparative study. Observations were made using light, fluorescence, differential interference contrast and scanning electron microscopy. Most of the characters of the ovary and ovule were typical of the family, except for the nucellar beak in Echinopsis aurea and E. haematantha, here reported for the first time in cacti. We found evidence suggesting that the stigmatic surface covered with multiseriate trichomes, the semi-closed style type and the pollen tube transmitting tract are conserved characters among species of Trichocereeae; this finding may be explained by a phylogenetic conservatism of the investigated genera of the tribe. We integrated the available information about structural and histological characters of the gynoecium in the family, taking into account the current phylogenetic context of the examined genera. Our results reinforce the significance of floral anatomical traits for the systematics of Cactaceae.
