Elucidation of Toxicity Pathways in Lung Epithelial Cells Induced by Silicon Dioxide Nanoparticles

Abstract: A study into the effects of amorphous nano-SiO2 particles on A549 lung epithelial cells was undertaken using proteomics to understand the interactions that occur and the biological consequences of exposure of lung to nanoparticles. Suitable conditions for treatment, where A549 cells remained viable for the exposure period, were established by following changes in cell morphology, flow cytometry, and MTT reduction. Label-free proteomics was used to estimate the relative level of proteins from their component tr… Show more

“…Many different types of NPs have also been found to affect cellular morphology [14,45], for instance by deregulation of the cytoskeletal architecture of the cells [46]. These effects have been found to occur at conditions at which the NPs did not cause any acute cytotoxicity [47].…”

“…Many studies also focus on the mechanisms of QDot toxicity and therefore employ CdTe core-only QDots [20,23,46,53], which are known to be much more toxic than the core-shell QDots. The current study focuses on QDots that have a great potential for biomedical use and seeks to assess the potential of these QDots for their intended purpose of fluorescence-based imaging at non-toxic conditions.…”

“…RAPGEF2 has been found to be critical for murine embryonic hematopoiesis and to be differentially expressed in pulmonary arterial hypertension (47,48). PHACTR1 plays a role in the reorganization and regulation of the actin cytoskeleton and appears to influence important vascular functions (49,50). MCPH1 is a gene that encodes a DNA damage response protein (23).…”

“…5 NPs may interact with tissue macromolecules and adversely affect cellular physiology. [6][7][8][9][10] Numerous studies have investigated the cytotoxic effects of SiO 2 and ZnO NPs, [11][12][13][14][15][16][17][18][19] but many controversies remain due to the physical and chemical properties of these NPs.…”
