Sugarcane mosaic virus (SCMV) is the main etiological agent of sugarcane mosaic disease, which affects sugarcane, maize and other economically important grass species. Despite the extensive characterization of quantitative trait loci controlling resistance to SCMV in maize, the genetic basis of this trait is largely unexplored in sugarcane. Here, a genome-wide association study was performed and machine learning coupled to feature selection was used for the genomic prediction of resistance to SCMV in a diverse panel of sugarcane accessions. This ultimately led to the identification of nine single nucleotide polymorphisms (SNPs) explaining up to 29.9% of the phenotypic variance and a 73-SNP set that predicted resistance with high accuracy, precision, recall, and F1 scores. Both marker sets were validated in additional sugarcane genotypes, in which the SNPs explained up to 23.6% of the phenotypic variation and predicted resistance with a maximum accuracy of 69.1%. Synteny analyses showed that the gene responsible for the major SCMV resistance in maize is probably absent in sugarcane, explaining why such a major resistance source is thus far unknown in this crop. Lastly, using sugarcane RNA sequencing data, markers associated with the resistance to SCMV in sugarcane were annotated and a gene coexpression network was constructed to identify the predicted biological processes involved in SCMV resistance. This allowed the identification of candidate resistance genes and confirmed the involvement of stress responses, photosynthesis and regulation of transcription and translation in the resistance to this virus. These results provide a viable marker-assisted breeding approach for sugarcane and identify target genes for future molecular studies on resistance to SCMV.

This study was conducted to investigate the effect of different in vitro techniques on elimination of dahlia mosaic virus. Meristems of Dahlia mosaic virus (DMV) infected dahlias, with 0.2-0.3 mm in length, were cultured on Murashige and Skoog (MS) medium. Based on DAS-ELISA, 100% of the survived plantlets by meristem culture were virus-free. DMV infected stem segments with two axillary buds were treated at three different levels of electric currents (15, 25 and 35 mA) and two time courses (10 and 20 min) in an electrophoresis tank and cultured on MS medium. The treatment of 35 mA for 20 min was the best for viral elimination from diseased dahlias by 85% followed by 25 mA for 20 min by 70%. Acyclovir and salicylic acid were the antivirals were used to eliminate DMV from the infected dahlias. For chemotherapy, nodes were cultured on MS media supplemented with antivirals at 0, 10, 20, 30, 40 and 50 mg/l for 20 and 30 days. The highest concentration of acyclovir (50 mg/l) for 30 days were the best treatments as its highest effectiveness on elimination ability (90.67%) but gave only 49% for plantlet survival. Whereas 40 mg/l acyclovir for 30 days gave elimination ability (80%) and plantlet survival (89.67%). The therapy containing salicylic acid at 40 mg/l for 30 days was the best as its highest effectiveness on DMV elimination ability (100%). But the concentration of salicylic acid at 30 mg/l for 30 days was the best treatment (chemotherapy efficiency= 49.21) as well as DMV elimination ability (75.33%) and plant survival (65.33%). The in vitro developed dahlia plantlets were tested for DMV-freeness using DAS-ELISA. The using therapies (electrotherapy and chemotherapy) can play a good role in virus elimination because they need the minimum period of time to regenerate fully developed healthy plants as long as 2-3 months.

Various RNA viral diseases on sugarcane result in yield loss and decreased sugar content. Breeding new varieties with virus resistance is the main goal of the sugarcane breeding program. Both single-stranded and double-stranded RNA viruses generated a double-stranded RNA replicative form (RF) during the replication cycle progress. While double-stranded RNA-specific ribonuclease (PAC1) encoded by the Pac1 gene (from Schizosaccharomyces pombe) can recognize and degrade double-stranded RNA specifically without any sequence, the expression of PAC1 in transgenic sugarcane may successfully develop virus-resistant sugarcane. In this research, we first expressed the PAC1 in prokaryotic cells. Then, double-stranded RNA RF of sugarcane's streak mosaic virus (SCSMV) was artificially synthesized. The degradation activity of the PAC1 was successfully tested by mixing the PAC1 protein and the double-stranded RNA RF. After that, the Pac1 gene was ligated to a plant expression vector and was then introduced into a virus-sensitive sugarcane cultivar by using Agrobacterium tumefaciens-mediated transformation method. Transgenic plants were challenged by inoculating with SCSMV. Results showed that although all the transgenic lines were infected by SCSMV, the mosaic symptoms that appeared on the leaves were significantly milder than that of the wild type. All transgenic shoots showed significantly lower viral loads and attained greater heights than wild-type shoots. This research provided a new pathway for breeding new varieties of sugarcane with virus resistance.
