We quantified associations between IgE and cancer in a meta-analysis and cohort study. Pubmed and Embase were searched to extract information using predefined inclusion criteria. In the Apolipoprotein MOrtality RISk (AMORIS) database, 24,820 persons had IgE measurements. Multivariate Cox proportional hazard models were used to analyze associations between IgE and cancer. Twenty-seven studies were reviewed from which seven case-control studies were included for analysis. The pooled relative risk (random effects model) was 0.97 (95% CI 0.86-1.09). Cell types of tumor origin (mesenchymal tissue or cells of the nervous system, lymphatic or hematopoietic tissue, and epithelium) modified the effect. In the AMORIS cohort, 862 persons developed cancer. Hazard ratios comparing quartiles of IgE were similar to the findings in the meta-analysis (HR 0.87 (95% CI 0.72-1.06); 0.94 (0.78-1.14); 0.90 (0.74-1.10) for the 2nd, 3rd, and 4th quartile compared to the 1st quartile), but there was no pattern by tumor origin. Both studies showed a weak inverse association between IgE and cancer, but a pattern by cancer type was only seen in the meta-analysis. Our findings suggest the need for prospective studies studying IgE and cancer. Measurements of IgE should be combined with other information, e.g., bio-banked samples containing other key immunological discriminators.

Circulating levels of eosinophil cationic protein (ECP), an eosinophil-specific granule protein, and peripheral eosinophils were determined in 56 patients with uremia. The patients were on a chronic hemodialysis or CAPD program or treated with diet only. They had normal counts of peripheral eosinophils but on average a threefold increase of the serum ECP concentrations compared with healthy subjects. The intracellular content of ECP in eosinophils isolated from 6 patients on maintenance hemodialysis was normal. The management or duration of the uremic condition did not influence the circulating ECP levels or eosinophil counts. Neither was any relation found between total IgE levels and peripheral eosinophils or serum ECP. The present results are compatible with an accelerated turnover of peripheral eosinophils in uremia. Such a proposed abnormal eosinophil homeostasis should demand an accelerated marrow eosinophil production, thereby explaining the previously reported marrow eosinophilia in azotemic patients.

In order to evaluate the possible preventive role of an anaphylactic condition on tumor development, C57BL/6J mice were immunized or hyperimmunized by the intraperitoneal route with egg albumin in alum before the chemical induction of fibrosarcoma. Preimmunization using a protocol which elicits an optimal IgE response produced: (1) a significant reduction of tumor incidence; (2) an increase of survival time, and (3) a decrease of tumor growth rate in animals with higher IgE titers (> 640). On the other hand, hyperimmunized mice, which were suppressed in their anti-egg albumin IgE response, showed no changes in tumor incidence and survival time when compared to controls. Our results suggest a suppressive effect on tumor development related to the ability of the animal to mount an IgE response to antigens unrelated to the tumor.
