Elevated Levels of Fragmented DNA Nucleosomes in Native and Activated Lymphocytes Indicate an Enhanced Sensitivity to Apoptosis in Sporadic Alzheimer’s Disease

Abstract: Apoptotic cell death is thought to be the most likely mechanism of cell death contributing to neurodegeneration in Alzheimer’s disease (AD). Here, we provide evidence that in sporadic AD cases the vulnerability of peripheral cells to undergo apoptosis is increased compared to non-demented elderly controls and, very importantly, to patients with subcortical vascular encephalopathy (SVE) as another, but demented control group. Quiescent ‘native’ and ‘activated’ lymphocytes from AD patients that were predisposed … Show more

“…Reductions of lymphocyte response to PHA and in vitro IL2 production are effects of repeated stress in rats (Batuman et al, 1990). The neurodegenerative disorder itself has proven effects on central and peripheral immune competent cells as well (Garibaldi and Zhang, 1999;Eckert et al, 2001). Recently, we could show that the CD4/CD8-ratio of Alzheimer's disease patients significantly correlates with the disease's severity (measured as cognitive decline).…”

“…Immune cell function is modulated by neuroendocrines as well as by neurotransmitters and vice versa (Hanisch et al, 1996;Carson and Sutcliffe, 1999). In addition, neurodegenerative diseases, such as dementia of Alzheimer type (DAT (Eckert et al, 1998(Eckert et al, , 2001Schindowski et al, 1999)), Down's Syndrome (DS (Park et al, 2000)) and the demyelating disease, multiple sclerosis (MS (Crucian et al, 1995a)) are accompanied by altered lymphocyte function and apoptosis. Chronic psychosocial and emotional stress is associated with declined and/or impaired immune function (Berkenbosch et al, 1991).…”

“…Low molecular weight DNA which is built during apoptosis is found only within the cell. As apoptosis proceeds in vitro, however, cells develop secondary in vitro necrosis which is characterized among other things by cell lysis and the appearance of DNA fragments in the supernatant (Watt et al, 1994;Eckert et al, 2000).…”

“…In addition, lymphocyte cell lines, e.g., the human Jurkat T-cell line, transfected with mutant PS1 (Passer et al, 1999) and lymphocytes from FAD patients bearing PS1 mutations (Parshad et al, 1996) show similar results in regard to increased vulnerability to DNA damage finally leading to accelerated cell death. Moreover, enhanced vulnerability to cell death and perturbed calcium homeostasis have also been described in peripheral lymphocytes from AD patients (Parshad et al, 1996;Eckert et al, 1997aEckert et al, , 1998Eckert et al, , 2000Gasparini et al, 1998).…”

“…In addition, lymphocyte cell lines (e.g., the human Jurkat T-cell line), transfected with mutant PS1 (Wolozin et al, 1998), and lymphocytes from FAD patients bearing PS1 mutations (Parshad et al, 1996) show similar results in regard to increased vulnerability to DNA damage leading to accelerated cell death. Moreover, enhanced vulnerability to cell death has also been described in peripheral lymphocytes from AD patients (Hartwig, 1995;Parshad et al, 1996;Eckert et al, 1998;Eckert et al, 2001a) but not in patients with vascular dementia (Hartwig, 1995;Eckert et al, 2001a). Oxidative stress mechanisms seem to influence this process (Gibson et al, 2000;Eckert et al, 2001a;Mecocci et al, 2002).…”
