Elevated immune response in Octopus rubescens under ocean acidification and warming conditions

Abstract: To study the effects of ocean warming and acidification on the immune response of octopuses, 24 Octopus rubescens were collected from Driftwood Park, Washington (48°09′48.9"N 122°38′14.1"W) April-July 2018. Following 3 weeks in elevated pCO 2 , elevated temperature, or the combination thereof, immunological parameters were measured including total hemocyte count, phagocytosis activity, superoxide production, and lysozyme activity. Increased pCO 2 elicited an increase in the number of circulating hemocytes, whi… Show more

“…Elevated CO 2 alters the molluscan immune response, with most research focusing on bivalves (Bibby et al, 2008;Liet al, 2015;Liu et al, 2016;Su et al, 2018;Wu et al, 2016)), though the immune response of an octopus was also affected by elevated CO 2 (Culler-Juarez & Onthank, 2021). Here, we found DE of genes that regulate immune signal transduction pathways and which are also implicated in the molluscan immune response (map4k5/3, syvn1-b, psenen , cbs) (Canesi et al, 2006;De Zoysa et al, 2010;Goodson et al, 2005;Salazar et al, 2015).…”

“…The temperature of the aquaria was not controlled and allowed to match the room temperature. Independent measurements of pH were taken of each aquarium daily using the m-cresol purple spectrophotometric method ( Dickson et al, 2007 ) as modified by Culler-Juarez and Onthank (2021) . Total pH values were calculated from absorbance values in R using the “specpH” function found the in OTools package version 22.08.01 ( https://github.com/KirtOnthank/Otools ).…”

“…After being held in aquaria for at least 1 day, octopuses were transferred to treatment tanks as previously described ( Culler-Juarez and Onthank, 2021 ). Each tank (113.5 L) was made using an insulated cooler with an overhead window in the lid to allow observation of octopus.…”
