Electrospray Quadrupole Travelling Wave Ion Mobility Time-of-Flight Mass Spectrometry for the Detection of Plasma Metabolome Changes Caused by Xanthohumol in Obese Zucker (fa/fa) Rats

Abstract: This study reports on the use of traveling wave ion mobility quadrupole time-of-flight (ToF) mass spectrometry for plasma metabolomics. Plasma metabolite profiles of obese Zucker fa/fa rats were obtained after the administration of different oral doses of Xanthohumol; a hop-derived dietary supplement. Liquid chromatography coupled data independent tandem mass spectrometry (LC-MSE) and LC-ion mobility spectrometry (IMS)-MSE acquisitions were conducted in both positive and negative modes using a Synapt G2 High D… Show more

“…Data were acquired in MS e mode with MS/MS fragmentation performed in the trap region. This technique enabled the best LC peak sampling by acquiring alternate low-energy/high-energy scans over the entire selected mass range, with no precursor selection in the quadrupole region (48). Low-energy scans were acquired at fixed 4-eV potential, and high-energy scans were acquired with an energy ramp from 25 to 45 eV.…”

“…Recently, approaches using another gas phase separation, ion mobility spectrometry (IMS) [ 66 ], has been gaining interest in metabolomics [ 67 , 68 , 69 , 70 , 71 , 72 , 73 ]. Indeed, IMS is a well-established post-ionization separation method based on size, shape, and charge performed on a millisecond timescale, which represents an intermediate timescale between chromatography (seconds) and high-resolution MS detection (microseconds).…”

“…Challenges due to the complexity of biological matrices could therefore be overcome using promising technologies like the traveling-wave ion mobility (TWIM) cell included in an orthogonal Q-TOF-MS. [22] This device enables separation of ionic species based on their size, charge and shape. [25][26][27] This approach has also recently been assessed in the context of chemical food safety applied to the specific and sensitive detection of small molecules like β-adrenergic agonists in complex biological matrices. Although this technology has mainly been investigated for large molecules [23] for the purpose of structural analyses, recent studies have presented interesting applications for small molecules detection in terms of separation efficiency [24] and specificity.…”

[25][26][27] This approach has also recently been assessed in the context of chemical food safety applied to the specific and sensitive detection of small molecules like β-adrenergic agonists in complex biological matrices. [28] The idea behind this study was first to compare LC and SFC separation performances of non-steroidal SARMs and secondly, to assess the potential of IMS-MS in relation with the analysis of these compounds when present at trace levels in biological matrices, but also to provide data related to enobosarm elimination and detectability in bovine urine.…”
