Electrophoretic study of the genome of human rotaviruses from Rio de Janeiro, São Paulo and Pará, Brazil

Abstract: Human rotaviruses from the states of Rio de Janeiro, São Paulo and Pará of Brazil were analysed by RNA electrophoresis. At least some bands characteristic of rotavirus double-stranded RNA were detected in 138 (86.8%) of 159 faecal samples in which the presence of rotavirus had been demonstrated by enzyme immunoassay. Of the RNA-positive samples, 18 (13.0%) were classified as subgroup 1, 94 (68.1%) as subgroup 2, and 26 (18.8%) could not be classified due to absence of visible bands 10 and 11. Subgroup 2 was mo… Show more

“…Cocirculation of different electropherotypes or replacement of one by another have been found in many parts of the world (Espejo et al 1980;Rodger et al 1981;Schnagl, Rodger & Holmes, 1981;Lourenco et al 1981;Flores et al 1982;Pereira et al 1983;Follett & Desselberger, 1983 a). At present it is not quite clear how these differences emerge and what their significance is.…”

“…The RNA of rotaviruses can be readily isolated and segments separated by electrophoresis on polyacrylamide gels (Kalica et al 1976(Kalica et al , 1978Espejo et al 1980;Schnagl, Rodger & Holmes, 1981;Rodger et al 1981;Lourenco et al 1981;Flores 210 E. A. C. FOLLETT AND OTHERS et al 1982;Pereira et al 1983;Follett & Desselberger, 1983a). RNA migration patterns ('electropherotypes', Rodger et al 1981, Schnagl, Rodger & Holmes, 1981; 'electrophoretypes', Thouless, Beards & Flewett, 1982) can thus be determined.…”

“…Considering the last population census carried out by Instituto Brasileiro de Geografia e Estatística (IBGE 2000), the sampling analyzed in relation to the infantile population was regarded as representative in both cities (95% confidence interval). All samples were primarily screened for rotavirus and adenovirus utilizing a combined enzyme immunoassay for Rotavirus A and adenovirus -EIARA - (Pereira et al 1985) and also by polyacrylamide gel electrophoresis -PAGE (Pereira et al 1983). Samples were also screened for calicivirus by RT-PCR using primers designed for the RNA polymerase region (Green et al 1995, Vinjé et al 1997.…”

“…For other types of rotavirus, polyacrilamide gel electrophoresis (PAGE) was used according to Pereira et al (1983) and the gels were stained with silver die (Herring et al 1982). Fecal samples were first submitted to extraction by the phenol /chloroform method, starting with a 20% fecal suspension.…”
