Electronic recording of lifetime locomotory activity patterns of adult medflies

Abstract: Age-specific and diurnal patterns of locomotory activity, can be considered as biomarkers of aging in model organisms and vary across the lifetime of individuals. Τhe Mediterranean fruit fly (medfly), Ceratitis capitata, is a commonly used model-species in studies regarding demography and aging. In the present study, we introduce a modification of the automated locomotory activity electronic device LAM25system (Locomotory Activity Monitor)–Trikinetics, commonly used in short time studies, to record the daily l… Show more

“…In this system, flies were individually kept in 32 glass tubes with 25 mm diameter and 125 mm length. A vinyl plastic stopper (CAP25-BLK- Vinyl Tube Cap-Trikinetics) was adjusted inside on the one end of the tube, which was an agar-based gel diet with sugar, yeast hydrolysate, agar, nipagin and water (4:1:0.2:0.1:20), for food and water provision [ 28 ]. The other end of the tube was covered with a piece of organdie to allow ventilation.…”

“…The protocol of Rodovitis et al (2022) was followed for both measuring detailed locomotor activity and maintaining the flies in the glass tubes. The day following adult emergence, we placed virgin females of the Mediterranean fruit fly laboratory strain "Benakio," which was maintained under laboratory conditions for more than 30 years (Kyritsis et al (2019)), in the tubes of the locomotory activity Monitor-LAM25 system and maintained them until death.…”
