Abstract: We summarize the history and recent development that has been made in the ECL detection of single entities.

“…Electrochemiluminescence (ECL) is the emission of light triggered by electrochemical reactions, which shows unique advantages, such as low background noise, high sensitivity, and maneuverability. Benefiting from the superiority of ECL and the specificity of immunoreaction, the ECL immunoassay (ECLIA) has become a powerful analytical tool for helping overcome the limitations of the enzyme-linked immunosorbent assay. However, the complex preparation of the antibody-tagged ECL probe and ECLIA platform limits their further development. Moreover, because of the low sensitivity of directly labeled antibodies, various tedious signal amplification strategies have been reported. In addition, the common sandwich-type ECLIA mode is usually an “on-electrode” state since a large number of labels are outside the Helmholtz plane surface, resulting in a less sensitive detection. Therefore, exploring a novel, convenient, and advanced multifunctional ECLIA probe to surmount these problems is still an active and continuous key aim for this issue.…”
