Electroejaculation and semen analysis in a male lowland gorilla,Gorilla gorilla gorilla

Abstract: ABSTRACT. Two electroejaculations and a testicular biopsy were performed on a lowland gorilla, Gorilla gorilla gorilla, to determine fertility. Sperm morphology showed 92.5 700 abnormal sperm which were of primary, or testicular, origin. Testicular biopsy revealed abnormally shaped nuclei in the later stages of spermiogenesis, supporting the semen analysis performed from the electroejaculations. The majority of abnormalities consisted of pyriform heads (23 ~o) and abnormal acrosomes (35 ~).

“…Studies of semen characteristics in aging nonhuman primates have been cross-sectional, often with single or short time-point measures and small sample sizes (Platz et al 1980;Tollner et al 1990;Schaffer et al 1992;VandeVoort et al 1993;Morrell 1997;Yeoman et al 1997;Ramesh et al 1998;Ji et al 2001). Some studies have focused on testosterone levels in male rhesus macaques, with somewhat contradictory results, similar to the human data (Mattison et al 2001;Black and Lane 2002;Mattison et al 2003;Roth et al 2004).…”

“…In mammals, including primate species, sperm pathologies are frequently associated with marked or subtle changes in sperm morphology (Chemes and Rawe 2003;Valle et al 2012). Sperm pathologies such as teratozoospermia are often observed in primates including humans (Platz et al 1980;Cui et al 1991;Baccetti et al 2002;Nakazato et al 2015). Traditionally, sperm morphology analyses have been carried out using subjective diagnostic methods to classify amorphous sperm heads, midpieces and tails.…”

“…Spermatozoa used in previous gorilla ART studies were collected either by electroejaculation using a rectal probe or from excised epididymides. Characteristically, gorilla semen collected by electrostimulation is of low motility and often has a high percentage of abnormal spermatozoa [Platz et al, 1980;Gould & Kling, 1982;Schaffer et al, 1989]. Therefore, in the present study, a young, reproductively naive gorilla male was conditioned to ejaculate by manual penile stimulation.…”
