Electrochemical Detection of miRNAs

Abstract: The recent progress made in the development of electrochemical methods for microRNA (miRNA) detection is presented. This progress is conceived to be largely due to the invention of novel assay methodologies and the use of various bioreagents and nanostructures. These enable a rigorous control over the sensing interface, provide enormous signal amplifications and single-base mismatch specificity. Femtomolar or even subfemtomolar detection limits were shown to be feasible by electrochemical assays. Thus electroc… Show more

“…In this study, the entire processes were done on the (DOTAP-DOPE) liposomal composition as the sandwich layer in GNP-SCPE 24 . Also to determine the effect of electrochemical changes on DOPE-DOTAP liposomes, each of the liposomes (DOPE and DOTAP), were used on the separate electrodes as control cases.…”

“…The initiation and progression of human cancers relies on alterations in miRs expression 2 . Circulating miRNAs seems as package in exosomes that derived from multivesicular bodies, or to be exported in the presence of RNA-binding proteins (i.e., Ago-2) or shed into micro vesicles within membrane blabbing 3 . According to recent literature on this field, circulating miRNAs have diagnostic and prognostic significance in cancer and many other diseases 4 .…”

“…Thus, many recent attempts at platform development (both uniplex and multiplex) have capitalized on adapting high-sensitivity transducers to novel, selectivity-enhanced recognition elements. As many excellent reviews (25,(61)(62)(63)(64)(65) have provided in-depth background on the various biosensor platforms and techniques, we limit our discussion to recent examples of this trend that utilize direct (unlabeled) miR detection methods.…”

“…The concept may be extended to the detection of proteins by using aptamer-based receptors as well as for a wide range of nucleic acids. In this latter case nucleic acid targets of a well-defined length, such as microRNAs [36], are preferred for readily quantifiable resistance changes during nanoparticle displacement assays. While in this study we used impedance spectroscopy to detect changes in the pore resistance, it would probably be feasible to use simple DC measurements instead.…”
