Electrocardiographic monitoring of cardiac transplants in mice

Abstract: In the murine cardiac model for vascularised transplantation graft function may be monitored by direct palpation, electrocardiography, and graft histology. To assess the merits of these methods, and to measure ischaemic damage, isografts and allografts were examined at regular intervals from 0 to 60 days after grafting. Heart transplants were vascularised from the abdominal great vessels, using microsurgical techniques, with ischaemic times of less than 60 min. Isografts showed no decrease in heart rate over 6… Show more

“…CBA/CaJ (H-2 k ) or BALB/cByJ (H-2 d ) donors depending on availability were used for heterotopic cardiac transplantation (13,14) into C57BL (H-2 b ) recipients that were either wild-type (CBA to C57BL/6J, n ϭ 11; BALB/c to C57BL/6J, n ϭ 8) or had targeted gene disruption in IFN-␥ (C57BL/6J, n ϭ 10) (15), IL-4 (C57BL/6J, n ϭ 10) (16), or IL-10 (C57BL/10j, n ϭ 10) (17). Isograft controls included IFN-␥ Ϫ / Ϫ ( n ϭ 7), IL-4 Ϫ / Ϫ ( n ϭ 6), and IL-10 Ϫ / Ϫ ( n ϭ 5).…”

“…Vascularized heterotopic heart grafts were anastomosed to the abdominal aorta and vena cava using microsurgical techniques 11 . Transplant function was evaluated by regular abdominal palpation and electrocardiogram (ECG) 12 . For skin grafting, donor tail skin was dissected into 5‐mm squares and secured with a Band‐aid to the graft bed on the recipient flank.…”

“…18 In another study, palpation of the heart graft gave an accurate measure of the pulse rate and correlated with electrical activity in acutely rejecting grafts, but in long surviving grafts, measurement of ventricular complex voltage showed myocardial damage that was not detectable by direct palpation. 17 In about 20% of cases with minimal intensity (þ1) by palpation, direct visualization showed no visible muscular contraction. 19 This problem occurs when misjudging passive movement of the graft by transmission of the aorta pulsation as graft beating.…”
