Electroacupuncture inhibits hippocampal neuronal apoptosis and improves cognitive dysfunction in mice with vascular dementia via the JNK signaling pathway

Abstract: Background: Electroacupuncture (EA) has been shown to reduce cognitive impairment in vascular dementia (VaD) patients. However, the mechanism of action remains unknown. Objective: The c-Jun N-terminal kinase (JNK) signaling pathway plays an important role in apoptosis. Herein, we focused on whether EA can inhibit apoptosis and alleviate cognitive impairment by regulating the JNK signaling pathway using a mouse model of VaD induced by modified bilateral common carotid artery occlusion (BCCAo). Methods: In exper… Show more

“…It is suggested that p53 and its downstream products may be another potential target for acupuncture to inhibit apoptosis and improve PSCI. EA can also upregulate the protein and mRNA expressions of Bcl-2, inhibit activator protein (AP)-1, p53, and Bax, alleviate the apoptosis of hippocampal cells in rats induced by modified bilateral common carotid artery occlusion (BCCAO), and improve their cognitive impairment, which may be related to the inhibition of JNK signaling pathway ( Liu et al, 2022 ). The process of apoptosis is highly controlled by various intracellular pathways, including nuclear factors-κB (NF-κB) Signal conduction.…”

“…It can also be damaged after stroke, resulting in cognitive dysfunction [ 29 ]. Research conducted by Yaru Liu and colleagues demonstrated that electroacupuncture may improve cognitive impairment caused by vascular dementia by inhibiting the JNK signaling pathway and reducing apoptosis in the hippocampus [ 30 ]. Additionally, through behavioral testing and proteomic analysis, Sa and colleagues found that electroacupuncture can improve the proteome in the hippocampal region of rats with cognitive impairment, which is associated with the upregulation of heat shock proteins in the brain via electroacupuncture treatment [ 31 ].…”

“…Bcl-2, Bax, and Casp3 can inhibit or promote the apoptosis of neurons; therefore, the regulation of neuronal apoptosis-related factors has a certain effect on VD control. Some studies have also proved that TCM therapy can play a role in the anti-apoptosis of nerve cells by regulating levels of Bcl-2, Bax, and other factors related to nerve apoptosis (13). Nevertheless, there is no clinical study about the influence of Shibing Xingnao Granules on factors related to nerve apoptosis.…”
