Electrical Lysis and RNA Extraction from Single Cells Fixed by Dithiobis(succinimidyl propionate)

Abstract: We present a microfluidic method for electrical lysis and RNA extraction from single fixed cells leveraging reversible cross-linker dithiobis(succinimidyl propionate) (DSP). Our microfluidic system captures a single DSP-fixed cell at a hydrodynamic trap, reverse-cross-links the DSP molecules on a chip with dithiothreitol, lyses the plasma membrane via electrical field, and extracts cytoplasmic RNA with isotachophoresis-aided nucleic acids extraction. All of the on-chip processes complete in less than 5 min. We… Show more

“…Hence, nucleic acids can be extracted and pre-concentrated in the presence of a leading electrolyte (LE) and a tailing electrolyte (TE) ( Fig. 7 A) ( Subramanian Parimalam et al, 2018 ). Another nucleic acid extraction method is ion concentration polarization (ICP)-based electrokinetic (EK) trapping, in which the selective transport of cations through a cation-selective membrane (CEM), under a DC electric field, induces an ion depletion zone with significantly amplified electric fields in the microchannel; this acts as an electric force barrier that prevents the passage of negatively charged species ( Ouyang et al, 2018 ).…”

“…Reagents to support stable entrapment or covalent crosslinking of analytes within cells with on-demand release also prolong potential sample storage times prior to assay and increase retrieval or extraction efficiency. In recent years, this nascent field has experienced an explosion of novel protocols for performing a wide array of single-cell bioanalytical assays including immunohistochemistry, proteomic measurements, and DNA/RNA-sequencing [68,[85][86][87][88][89]. The following sections will serve to characterize the most significant recent developments of innovative single-cell preservation protocols.…”

“…4A) [91]. Reagents such as homobifunctional NHS-ester crosslinking reagents are more selective and less reactive than aldehydes; these reagents have been used to successfully preserve single cells while maintaining cellular structure and surrounding membranes, facilitating RNA-seq on single cells stored at 4°C up to 3 days post-fixation [89,96].…”
