Electrical and histological remodeling of the pulmonary vein in 2K1C hypertensive rats: Indication of initiation and maintenance of atrial fibrillation

Abstract: ObjectiveHypertension is a significant risk factor for atrial fibrillation (AF). The role of pulmonary vein (PV) remodeling in the mechanistic association between hypertension and AF is not definitive. In this study, we aimed to identify changes in the electrophysiology and histology in PVs in two-kidney, one-clip (2K1C) hypertensive rats.MethodsFifty male Sprague-Dawley rats were classified into the 2K1C and sham-operated groups. The systolic blood pressure was measured every 2 weeks. The left atrial diameter… Show more

“…At the same time, Western blotting indicated the expression of Kir2.1 in subgroup R-NAA was significantly lower, which may be related to the denaturation and remodeling of Kir2.1 in the atrial myocardium after hypothermic IR. These findings agree well with that of previously reported by Xia et al, 24 suggesting that downregulation of Kir2.1 may be related to the prolongation of atrial repolarization duration in rats with atrial arrhythmia.…”

“…Our findings are consistent with prior studies in rat models of hypertension, where reduced refractoriness and SCN5A (sodium channel protein type 5 alpha subunit) expression, as well as heterogeneous conduction, have been demonstrated. 21,22 Nonetheless, there are conflicting results for atrial electrophysiological remodeling seen in other hypertension models, likely caused by differences in methods (eg, dose or duration of Ang II infusion), method of generating hypertension, and concomitant structural remodeling.…”

“…For instance, thicker or longer sleeves were more frequent AF [23]. Recently, it was reported that more synthetic-like SMCs presenting in the pulmonary vein of hypertensive rats were more likely subjected to AF [14,24]. However, it remains to be elucidated whether these synthetic-like SMCs directly regulate bioactivities of cardiomyocytes.…”
