“…This increased elastinolysis was also shown in experimental PH induced by chronic hypoxia [59]. JACOB et al [60] and HORNEBECK et al [61] showed that a serine elastase was produced by aortic SMCs and associated with atherosclerosis, which was further characterised by ZHU et al [62] as EVE, and found in PA-SMC [62]. This elastase, like the polymorphonuclear neutrophil serine elastase, is inhibited by a 1 -antitrypsin/a 1 -proteinase inhibitor (a 1 -PI), a 2 -macroglobulin, elafin and by some synthetic inhibitors.…”

“…Elastin fragmentation has been shown to be an early pulmonary vascular abnormality in patients with congenital heart defects and PAH [2]. Elastinolytic activities have also been ascribed to endogenous vascular elastase [24,25]. In addition, elastinolytic properties of MMP-2 should also be taken into account [26].…”

“…These findings, in addition to those obtained in PAMs from smokers or in BAL cell extracts, suggest that three distinct types of elastase activity are liable to be present in PAMs. The first is a neutral metalloelastase-Iike activity, capable of hydrolysing SLAPN between but P 1 and P 2 Ala-Ala peptide bond but exhibiting no solubilizing elastin potential; it might be analogous to the endopeptidase found largely in cells such as lung cells and skin fibroblasts [50,51], bone marrow cells [52] or neutrophils [53] and in body fluids such as serum [33,[54][55][56][57] rheumatoid synovial fluid [58]; the complete processing of SLAPN cleavage would require the combined action of this endopeptidase and an aminopeptidase. The second type of PAM elastase is a metalloelastase capable of hydrolysing 3 Helastin; it shares several similarities with proteoglycanase also designated as stromelysin [28,39].…”
