Elaidic Acid, a Trans-Fatty Acid, Enhances the Metastasis of Colorectal Cancer Cells

Abstract: The effects of trans-fatty acids (TFAs) on cardiovascular disorders have been extensively studied, and the effect of TFAs on cancers has recently been recognized. This study examined the effects of elaidic acid (EA), a TFA, on colorectal cancer (CRC) progression. We demonstrated that EA enhanced the growth, survival, and invasion of the CRC cell lines, CT26, and HT29. Tumor growth and metastasis in the lung, liver, and peritoneum were significantly more enhanced in CRC cells treated with EA than those treated … Show more

“…LA induces senescence in cultured CRC cells and dormancy in CRC cells inoculated into mice (11). Conversely, EA increases the proliferation of cells in spheres via the activation of epidermal growth factor receptor and the Wnt signaling pathway, and the upregulation of stem cell markers (16,17). EA also induces epithelial-mesenchymal transition and enhances tumor growth and metastasis (16,17).…”

“…The cells (1x10 4 per well) were seeded in a 12-well dish. Cell growth was assessed using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay or by counting the number of cells with an Autocytometer (Sysmex, Kobe, Japan) after 48 h, as described previously (13 For analyzing the metastasis to the liver, lung, and peritoneum, cancer cells (1x10 6 cells) were inoculated into the spleen, the tail vein, or peritoneal cavity, respectively.…”

“…We have previously reported the pro-tumoral effects of elaidic acid (EA), a major trans fatty acid (TFA) (4,5). EA is a long-chain fatty acid (LCFA) containing 18 carbon chains in a trans configuration, and is a structural isomer of oleic acid (OA).…”

“…TFA are associated with carcinogenesis [18][19][20] . Moreover, cancer metastasis [21,22] and drug resistance [23] are promoted by elaidic acid (EA), a major component of dietary TFA [24] , through the enhancement of stemness and epithelial-mesenchymal transition [22,23] .…”
