Einführung

State-of-the-art of process development, scale-up and design for downstream processes is described for complex mixtures in biotechnology. The focus is on fermentation broth and plant-based extracts to derive methodological arguments. Academic and industrial approaches are discussed with an assessment based on some process relevant criteria. Examples of monoclonal antibody, insulin and penicillin production processes are chosen to develop a sound argumentative basis.Demands of total process integration, accuracy and sensitivity of phenomena, resulting e.g. in miniaturization of laboratory experiments and linked with necessary in-depth modeling are developed and implemented. Specific tasks of unit operation are displayed and their integration into an efficient process sequence is demonstrated. Based on that, further research needs are pointed out and corresponding activities are identified. For instance, the transfer of well established methods in chemical engineering like process modeling in combination with laboratory scale experiments and final miniplant validation should be followed consequently also in biotechnology applications. Some drawbacks in academic education and industrial training as well as organizational structures are discussed to help address this issue.

Die numerische Strömungsmechanik gewann in den letzten Jahrzehnten zunehmend an Bedeutung. Verantwortlich dafür war zum einen die kontinuierlich steigende Leistung der Rechner, zum anderen die Entwicklung mathematisch‐physikalischer Rechenmodelle und numerischer Lösungsalgorithmen, die eine realitätsnahe Simulation komplexer Strömungsvorgänge erlauben. In dieser Arbeit wird die Volume‐of‐Fluid‐Methode eingesetzt, um eine Zweiphasenströmung in einem Rohr zu simulieren. Die Effekte des Kontaktwinkels, der Temperatur und des Massenstroms der Gasphase auf die Form der flüssigen Phase werden untersucht.

Simulation of Internal Recycling of Adsorption Heat with Phase Change MaterialsPhase change materials are able to store heat through a liquefaction phase change at a predetermined temperature. When using amine-functionalised adsorbents for CO 2 removal, the excess adsorption heat leads to a decrease in equilibrium capacity, which can thus be mitigated by storing the heat for adsorbent regeneration during the desorption step in a vacuum swing adsorption. This integration diminishes the temperature variations and renders the process more efficient, whereat the exact integration strategy has a strong influence on the overall efficiency of the phase change material performance.
