EGFR-mediated apoptosis via STAT3

Abstract: The Epidermal Growth Factor Receptor (EGFR) is a cell surface receptor with primary implications in cell growth in both normal and malignant tissue. Paradoxically, cell lines that hyperexpress the EGFR have been documented to undergo receptor-mediated apoptosis. The underlying mechanism by which EGF-induced apoptosis occurs however remains inexplicit. In an attempt to identify this mechanism, we assessed downstream effectors of EGFR in MDA-MB-468 cells during conditions of EGF-induced apoptosis. The effector a… Show more

“…In order to assess whether the chemosensitizing effects of β-caryophyllene (50 µM) towards doxorubicin were dependent on STAT3, we also tested the substances and their combination in human prostatic PC3 cells ( Figure S4), known to be lacking in the expression of STAT3 [66,67]. Under our experimental conditions, the cytotoxic effect of doxorubicin was slightly increased by β-caryophyllene, although no statistical difference was found in the IC 50 values (48.4, C.L.…”

“…However, when overexpressed, STAT3 promoted apoptosis through induction of the expression of caspase-3 [169]. Furthermore, mitoSTAT3 contribute to anti-apoptotic functions in tumour cells by increasing the MMP and inhibiting the opening of the MPTP to release Ca 2+ and cytochrome c into cytosol which would lead to intrinsic apoptosis as demonstrated in human esophageal squamous cell carcinoma (ESCC) [170,171]. Therefore, the role of STAT3 in apoptosis depends not only on the localization of STAT3, but its levels.…”

“…Apoptosis is also part of the regular turnover of the SG, when exhausted saliva-producing acinar cells die, and are replaced with new cells generated from progenitor cells. Enhanced EGF signaling, critical to epithelial cell and SGPC homeostasis, and also involved with STAT3-based cytokine signaling, can also result in apoptosis [93]. This provides an additional explanation for enhanced apoptotic levels observed in the epithelium of pSS patients, and also further confirms the complicity of STAT3 in a multitude of salient pathways.…”
