EGFL7 drives the evolution of resistance to EGFR inhibitors in lung cancer by activating NOTCH signaling

Abstract: Accumulating evidence supports evolutionary trait of drug resistance. Like resilience in other systems, most tumor cells experience drug-tolerant state before full resistance acquired. However, the underlying mechanism is still poorly understood. Here, we identify that EGF like domain multiple 7 (EGFL7) is a responsive gene to epidermal growth factor receptor (EGFR) kinase inhibition during a period when tumors are decimated. Moreover, our data reveal that the adaptive increase of EGFL7 during this process is … Show more

“…On the other hand, the CAV1 gene, notorious for bolstering lung cancer cell proliferation, manifests heightened expression during pathological conditions [56]. Other markers, such as HSPB1, EGFL7, and PPIA, while not overtly differentially expressed, resonate strongly with their established biological significance [57,58,59]. Figure 3(b) serves as an emblem of scSniper's autoencoder module's finesse in sculpting biologically coherent cell embeddings, facilitating a precise delineation of diverse cell type clusters.…”

“…Additionally, there is a crosstalk between Notch signaling and other pathways, such as TGF-β and EGFR. Activated Notch pathway leads to an expansion of TGF-β-induced smad2/3 signaling and EGFR inhibitors resistance [ 14 , 76 ]. Notably, the pathogenic role of the Notch pathway appears to be tumor type dependent.…”
