EGCG synergizes the therapeutic effect of cisplatin and oxaliplatin through autophagic pathway in human colorectal cancer cells

Abstract: Application of the platinum-based chemotherapy for colorectal cancer is restricted due to its severe cytotoxic effects. In this study we used synergistic strategies by combining (-)-Epigallocatechin gallate (EGCG) with cisplatin or oxaliplatin to minimize the ill effects of platinum-based therapy. MTS assay was used to examine the effect of EGCG, cisplatin and oxaliplatin on the proliferation of human colorectal cancer DLD-1 and HT-29 cells. Autophagic process was evaluated by detection of LC3-II protein, auto… Show more

“…At the same time, EGCG promoted the growth inhibition effect of antitumor drugs through autophagy regulation via activation of pro-apoptosis or pro-survival autophagy in cancer cells [3,29,30]. Based on the synergistic effect of EGCG and DOX on osteosarcoma growth inhibition, in second part of this study, it was confirmed that EGCG inhibited DOX induced pro-survival autophagy which would be one of the important molecular mechanism of synergistic effect of EGCG and DOX on osteosarcoma.…”

“…EGCG enhanced the effect of cisplatin and oxaliplatin-induced autophagy in human colorectal cancer DLD-1 and HT-29 cells, as characterized by the accumulation of LC3B-II protein, the increase of acidic vesicular organelles, and the formation of autophagosome, as described in [136]. Exposure of squamous cell carcinoma SSC-4 cells to EGCG induced activation of apoptosis and autophagy, associated with upregulation of BAD, BAK, FAS, IGF1R, WNT11, and ZEB1 protein levels, and downregulation of CASP8, MYC, and TP53 proteins, as indicated in [137].…”

“…84,89,90 For instance, platinum-based therapy and epigallocatechin gallate-mediated cell death may also depend on the generation of ROS either directly or indirectly. 91,92 A recent study suggested that Cis induces autophagy in HeLa cells via upregulating the formation of autophagic vesicles, promoting the conversion of LCI to LCII and increasing the expression of the autophagy-related ATG7 gene. 82 Our findings provide evidence that the autophagy induced by Cis …”
