Efficient screening for catalytic antibodies using a short transition‐state analog and detailed characterization of selected antibodies

Abstract: One of the major obstacles to acquiring catalytic antibodies is that it requires labor-intensive procedures to select catalytic antibodies from huge repertories of antibodies. Here, we selected potential catalytic Abs by utilizing their affinity towards a short transition-state analog which contained only the transition-state structural element, and evaluated in detail its efficiency to enrich catalytic Abs. Hybridoma supernatants elicited against a phosphonate derivative, the TSA1, were screened by a three-st… Show more

“…Samples were eluted with 0.1% trifluoroacetic acidacetonitrile (40:60) at a flow rate of 1.0 ml/min, and the results were monitored at 254 nm. The amounts of the substrate and product were calculated from their peak areas on the HPLC chromatogram, as described previously (27). The kinetic parameters k cat and K m were determined by fitting data to a MichaelisMenten equation by using the program KALEIDAGRAPH (Synergy Software, Reading, Pa.).…”

“…This study was motivated by previous findings of naturally occurring catAbs in patients with autoimmune diseases, such as asthma (14) or systemic lupus erythematosus (SLE) (15). Using a different phosphonate hapten, we have confirmed that catAbs could be recovered at higher incidence from the MRL/lpr mice than from the conventionally used normal mouse strains (16,17).…”

“…Some research groups isolated anti-growth hormone and antiprolactin antibodies in serum [68,69]. Further research showed that autoantibodies found in the serum of patients show a cross-reaction with neuron-specific enolase, which is restricted to neural tissue.…”
