The effects of rolC on morphogenesis of Chrysanthemum morifolium Ramat. 'Ogura-nishiki' were studied. The ray flowerets in the rolC-transformed plants were shorter than those in wild-type plants, but the transformation enhanced outgrowth of lateral shoots and reduced internode elongation. Studies on adventitious shoot and root formation from leaf segments revealed that the rolC transgene led to reduced root formation but enhanced adventitious bud formation. This enhanced cytokinin-like activity was consistent with phenotypes observed in the rolC-transformed plants.

We give an overview of the advances of an Agrobacterium-mediated transformation system, clarifying its problems and their solutions, and then show the latest version of our transformation system and examples of the introduction of agronomically important traits into chrysanthemums. Typical problems with the Agrobacterium-mediated transformation in chrysanthemum include low transformation e ciency, high chimerism and cultivar speci city. Using a co-cultivation medium containing acetosyringone and casamino acids for high transformation e ciency and an antibioticselection step for transgenic calli before plant regeneration to eliminate the chimerism, we established an e cient and stable transformation system for chrysanthemum. In addition, this system was used to successfully introduce useful agronomical traits, such as insect resistance and new ower color, into chrysanthemums. ese traits have been stably and highly expressed to confer the expected characteristics upon the transgenic chrysanthemums. Before applying a eld trial of the genetically modi ed (GM) chrysanthemums, male and female sterility were introduced into the transformants to exclude the transgene ow from the GM plants to their wild relatives. So far, using RNAi technology, some of the transgenic chrysanthemums have displayed complete male sterility with very weak female fertility.

Double-stranded RNA-specific Ribonuclease Confers Tolerance against <i>Chrysanthemum Stunt Viroid</i> and <i>Tomato Spotted Wilt Virus</i> in Transgenic Chrysanthemum Plants

We produced transgenic chrysanthemum (Dendranthema grandiflora) plants expressing double-stranded RNA-specific ribonuclease gene (pac1) derived from Schizosaccharomyces pombe using an Agrobacteriummediated transformation method. Three transgenic lines stably expressing Pac1 protein were selected. After inoculation with chrysanthemum stunt viroid (CSVd), these lines showed a decreased frequency of infection, less accumulation of viroid, and attenuation of growth retardance compared with control plants. They also showed a significantly lower infection frequency against tomato spotted wilt virus (TSWV) than in control plants. Hybrid plants between one transgenic line and a wild chrysanthemum species, D. pacificum, were produced to test their susceptibility against TSWV infection. TSWV infected all the plants of the wild species resulting in a high mortality, whereas the hybrid plants expressing Pac1 protein showed either complete resistance or high susceptibility. These results suggest the presence of an endogenous tolerance gene against TSWV in the cultivar used for transformation. The virus infection was suppressed in transgenic plants, indicating that the commercial use of this transgene would limit the spread of viruses to wild populations of Dendranthema. This is the first study to demonstrate that a single tolerance gene in transgenic plants can attenuate both viroid and virus diseases.
