Efficient Generation of Trunk Neural Crest and Sympathetic Neurons from Human Pluripotent Stem Cells Via a Neuromesodermal Axial Progenitor Intermediate

Abstract: The neural crest (NC) is a multipotent embryonic cell population that generates various cell types in an axial position-dependent manner. Cranial NC cells, give rise to mesoectodermal derivatives, melanocytes, neurons and glia whereas the vagal NC generates the enteric nervous system and trunk NC cells produce sympathetic neurons and neuroendocrine cells. An attractive approach for studying human NC biology and modelling NC-associated developmental disorders (neurocristopathies) involves the in vitro productio… Show more

“…In another key study, Muotri and colleagues reported a protocol where cortical organoids were induced with electrophysiologically active neurons with glutamatergic and GABAergic signaling ( Trujillo et al, 2018 ). We designed our protocol based on these existing reports and made use of selected hormones, such as human insulin, transferrin, and progesterone, which have been found to be protective or support the survival and differentiation of dopaminergic neurons and neural crest lineage cells ( Ciarlo et al, 2017 ; Frith and Tsakiridis, 2019 ). Of note, our findings suggest that the use of an orbit shaker might have contributed to quick induction of hBSOs, through the achievement of an ideal concentration gradient of growth factors.…”

“…Differentiation of NCC was carried out according to previously described procedures with minor modifications [27]. hPSCs were plated at 4 × 10 4 cells/cm 2 on plates coated with vitronectin in Essential 6 medium supplemented with CHIR99021 (1.4 μM), FGF2 (100 ng/ml), BMP4 (20 ng/ml), and SB431542 (10 μM, Peprotech).…”
