Efficient Assembly of Photosystem II in Chlamydomonas reinhardtii Requires Alb3.1p, a Homolog of Arabidopsis ALBINO3

Abstract: Alb3 homologs Oxa1 and YidC have been shown to be required for the integration of newly synthesized proteins into membranes. Here, we show that although Alb3.1p is not required for integration of the plastid-encoded photosystem II core subunit D1 into the thylakoid membrane of Chlamydomonas reinhardtii, the insertion of D1 into functional photosystem II complexes is retarded in the Alb3.1 deletion mutant ac29. Alb3.1p is associated with D1 upon its insertion into the membrane, indicating that Alb3.1p is essent… Show more

“…To test whether they correspond to known photosynthetic complexes, the BN gel blots were probed with antibodies against a subunit of each complex. This revealed that the 300-and 600-kD complexes correspond to the reaction center core PSII monomer and dimer, respectively, because they react with D2 antibodies and are absent in FuD7 ( Figure 4A, aD2) (Ossenbuhl et al, 2004). A complex with slower migration than the PSII dimer is detectable at ;700 kD with PsaA antiserum, corresponding most likely to PSI-LHCI ( Figure 4A, aPsaA).…”

“…It is absent in F15 and strongly reduced in ac29. Instead, a new complex is detectable in ac29 at 300 kD corresponding to PSI without LHCI (Ossenbuhl et al, 2004). This is attributable to the considerable decrease of LHCs in ac29 (Bellafiore et al, 2002;Ossenbuhl et al, 2004).…”

“…Two Alb3 proteins (Alb3.1 and Alb3.2) are present in Chlamydomonas, whereas only one Alb3 protein is present in Arabidopsis. Alb3.1 of Chlamydomonas is involved mainly in the insertion of the light-harvesting complex [22], and it is required for the insertion of D1 into functional PSII [23]. Alb3.2 interacts with Alb3.1 and the reaction centre polypeptides of PSI and PSII.…”

“…Psb29 and the immonophilin FKBP20-2 are required for efficient assembly of PSII supercomplexes (Keren et al, 2005;Lima et al, 2006;Fu et al, 2007). LPA2 and LPA3 cooperate with each other and interact with ALB3 in assisting incorporation and stabilization of CP43 (Ossenbühl et al, 2004;Cai et al, 2010). Psb28 was suggested to be important for the synthesis of chlorophylls and/or apoproteins of the inner antenna subunit CP47 (Dobáková et al, 2009).…”
