Over the last decade, capillary electrophoresis gained tremendous importance, because it became an indispensible tool for the quality control of biologics, e.g. therapeutic antibodies. Consequently, there has been a continuous development within the CE market. Microchip techniques have been established in the last years. Further trends are complete solutions for specific applications by the usage of reagent kits. Step by step instructions and facilitated handling of the instruments are becoming more common. This work focuses on the sized-based protein analysis with CE-SDS. The instruments CE 7100 by Agilent Technologies, LabChip® GXII Touch HT by PerkinElmer, Maurice S. by Protein Simple and PrinCE NextI870 by Prince Technologies have been evaluated, mainly analyzing protein mixtures of different molecular weights in long series. Published data of the PA 800 plus by SCIEX are also included in the tabled results. Precision, reliability, flexibility, and speed have been identified as the most important performance parameters, others such as resolution, sensitivity, linearity, ease of use and sustainability have also been considered. All tested instruments have shown an excellent performance. Depending on application and necessities, each user can find the most appropriate one.

Резюме Введение. Квалификация оборудования является одним из краеугольных камней обеспечения качества лекарственных средств, которые обеспечивают постоянство метрологических характеристик. Необходимость формирования отечественных руководств к проведению процесса квалификации средств измерений и гармонизация его с европейскими и американскими требованиями, позволит повысить качество лекарственных препаратов, выпускаемых в Российской Федерации, и повысит их конкурентоспособность на мировом фармацевтическом рынке. Текст. Цель настоящего обзора-анализ отечественной законодательной базы, которая регламентирует постоянство метрологических характеристик средств измерений, применяемых в фармацевтической промышленности. В статье приводятся данные, характеризующие особенности подтверждения соответствие оборудования метрологическим требованиям с точки зрения различных нормативных документов, действующих на территории Российской Федерации. Заключение. В статье нами продемонстрирована неоднозначность толкований отечественного законодательства и Европейских рекомендаций по качеству в сфере метрологических требований к средствам измерений. Разработка отечественного руководства, гармонизирующего метрологические требования к оборудованию, позволит сфокусироваться на результате и качестве лекарственных средств, а также устранит неоднозначность толкований отечественной законодательной базы. Ключевые слова: лабораторное оборудование, аналитическое оборудование, измерительная техника, квалификация оборудования, контроль качества лекарственных средств, Государственная фармакопея, стандарты качества лекарственных средств, метрологические требования, калибровка оборудования. Fast investigations from biological matrices using CE – Test of a blood–brain barrier model

In order to adopt a general workflow for complex biological matrices with respect to a new blood-brain barrier (BBB) model, a micellar electrokinetic chromatography method has been developed. The cells forming the BBB have been cultivated in a special cell growth medium in which six drugs (acetaminophen, caffeine, carbamazepine, cimetidine, indometacin and propranolol) have been dissolved and tested for their penetration properties. The results showed good to very good accordance to the reference values. Samples were directly injected onto the capillary without any pretreatment (fused silica capillary, id: 50 μm, L: 48 cm, l: 40 cm). After method development, separations were carried out using a 60 mM borate buffer containing 200 mM of SDS at 30 kV, leading to an analysis time of less than 10 min. Between two runs the capillary was rinsed with a mixture of equal parts of running buffer and isopropanol (70% v/v), which proved to be very effective to remove matrix compounds. An appropriate choice of the detection wavelength (220 or 254 nm) could avoid major interferences between analytes and matrix. The typical RSD% for migration times was approximately 2%, for peak areas, it ranged from 2 to 6%, which was very well acceptable for the generic method used in this study and the low concentrations investigated. The LODs ranged from 10 to 30 ng/mL.
