Canadian public oilseed breeding programs have played a leading role in the development of canola-quality Brassica rapa oilseed cultivars, but the genetic variability of the cultivars developed over the past 60 yr is poorly understood. Simple sequence repeat (SSR) markers were applied to assess the genetic variability of 294 plants representing one landrace introduced from Poland ca. 1940 and nine Canadian elite B. rapa cultivars released by Agriculture and Agri-Food Canada since 1964. Application of 18 SSR primer pairs detected 27 likely loci on five or more linkage groups and 123 polymorphic alleles. The allelic frequencies ranged from 0.003 to 0.997 and averaged 0.262. The estimates of mean heterozygosity for these cultivars ranged from 0.126 to 0.197 and averaged 0.165. Significant decreases of SSR alleles and average dissimilarities were observed over the 60 yr of breeding effort. The proportion of total SSR variation residing among the cultivars was 10.4%; between high vs. low erucic acid cultivars 5% and between high-vs. low-glucosinate cultivars 5%. Pairwise genetic differentiation ranged from 0.025 to 0.184 and averaged 0.104. Genetic clustering of these cultivars revealed AC Sunbeam to be a genetically unique cultivar, and two distinct groups of the other nine cultivars were separated by high-vs. low-glucosinate content. Several distinct genotypes, largely derived from the landrace Polish, were identified. These findings are useful for broadening the genetic base of elite B. rapa germplasm, selecting genetically diverse genotypes for synthetic and hybrid combinations, and conserving this germplasm. The objective of this study was to quantify the molecular diversity and to determine the genetic relationships among Secale spp. and among cultivars of Secale cereale using RAPDs, ISSRs and sequence analysis of six exons of ScMATE1 gene. Thirteen ryes (cultivated and wild) were genotyped using 21 RAPD and 16 ISSR primers. A total of 435 markers (242 RAPDs and 193 ISSRs) were obtained, with 293 being polymorphic (146 RAPDs and 147 ISSRs). Two RAPD and nine ISSR primers generated more than 80% of polymorphism. The ISSR markers were more polymorphic and informative than RAPDs. Further, 69% of the ISSR primers selected achieved at least 70% of DNA polymorphism. The study of six exons of the ScMATE1 gene also demonstrated a high genetic variability that subsists in Secale genus. One difference observed in exon 1 sequences from S. vavilovii seems to be correlated with Al sensitivity in this species. The genetic relationships obtained using RAPDs, ISSRs and exons of ScMATE1 gene were similar. S. ancestrale, S. kuprijanovii and S. cereale were grouped in the same cluster and S. segetale was in another cluster. This study was conducted in order to identify quantitative trait loci (QTLs) for the in vitro culture response of winter rye (Secale cereale L.) immature embryos and immature inflorescences. A genetic linkage map comprising 67 SSRs, 9 ISSRs, 13 SAMPLs, 7 RAPDs, 2 SCARs and one EST marker was created based on the analyses of 102 recombinant inbred lines from the cross between lines L318 (which has a good response in tissue cultures) and L9 (which is unable to regenerate plants from somatic tissues and anthers). The map spans 979.2 cM, and the average distance between markers is 9.9 cM. Two characteristics were evaluated: callus induction (CI) and somatic embryogenesis ability (SE). They were expressed as the percentage of immature embryos/inflorescences producing callus (designated ECI/ICI) and the percentage of explants producing somatic embryos (ESE/ISE). All the analysed traits showed continuous variation in the mapping population but a non-normal frequency distribution. We identified nine putative QTLs controlling the tissue culture response of rye, explaining up to 41.6% of the total phenotypic variation: two QTLs for ECI — eci-1, eci-2; 4 for ESE — ece-1, ese-2, ese-3, ese-4; 2 for ICI — ici-1, ici2; and 1 for ISE — ise-1. They were detected on chromosomes 1R, 4R, 5R, 6R and 7R.
