Efficacy of two formulations of afoxolaner (NexGard® and NexGard Spectra®) for the treatment of generalised demodicosis in dogs, in veterinary dermatology referral centers in Europe

Abstract: BackgroundA multi-centre field trial was conducted to evaluate the efficacy of afoxolaner based chewables (NexGard® or NexGard Spectra®) for the treatment of generalised demodicosis caused by Demodex canis in dogs under field conditions in France, Italy and Poland.MethodsClient-owned dogs, diagnosed positive for Demodex mites by pre-treatment skin scrapings and presenting clinical signs of generalised demodicosis were included. Dogs were orally treated with afoxolaner three times at monthly intervals. Of the 5… Show more

“…The only dog needing administration every two weeks was on immunosuppressive therapy for pemphigus foliaceus that became mite‐positive when the interval was increased to four weeks, but remained mite‐negative when afoxolaner was administered every two weeks . In a further study, 50 dogs with generalized demodicosis were treated with afoxolaner alone (Nexgard ® ) or combined with milbemycin (Nexgard Spectra ® ) with 2.5–2.7 mg/kg once monthly and the number of mites on skin scrapings was reduced by 87.6%, 96.5% and 98.1% on days 28, 56 and 84, respectively, and 36 of 50 dogs had negative skin scrapings after three months A more recent study looked at afoxolaner combined with milbemycin (Nexgard Spectra ® ) at the dosage of 2.5–6.3 mg/kg p.o. every four weeks in dogs with juvenile‐onset (n = 4) and adult‐onset (n = 11) generalized demodicosis with a variety of clinical lesions.…”

“…Where necessary (e.g. long or medium-haired dogs), hair was removed at the area to be scraped, and the skin was rmly squeezed prior to and during scraping to eject mites from hair follicles [2,14,15]. Scrapings were made in the direction of hair growth with a blade or spatula, covered with mineral oil, until capillary oozing was observed.…”

“…The efficacy of the treatment of demodicosis with isoxazolines has been described (Six et al, 2016;Snyder et al, 2017;Lebon et al, 2018). The oral treatment of this disease with fluralaner was effective as observed by other authors (Fourie et al, 2015;Bezerra et al, 2017;Duangkaew et al, 2018) with a single-dose administration.…”
