Efficacy of Liposomal Cytarabine in the Treatment of Leptomeningeal Metastasis of Breast Cancer

Abstract: Background: Breast cancer is one of the most frequent causes of leptomeningeal metastasis (LM) among solid tumors in adults. The prognosis of patients with LM is poor. A treatment option is the intrathecal administration of liposomal cytarabine. Case Reports: The first case represents a 53-year-old woman with LM of breast cancer. A neurological response was achieved shortly after the start of the multimodal treatment including intrathecal liposomal cytarabine. The response duration reached 79 days. The second … Show more

“…However, cytarabine has not yet been investigated in cervical cancer, nor the rs5030743 and rs1130609 of RRM2. Although cytarabine differs from gemcitabine in several important respects ( 86 ), for instance, gemcitabine differs structurally from cytarabine by a fluorine group substituted at position 2′ on the furanose ring ( 87 ), gemcitabine ( 88 90 ) and cytarabine ( 91 , 92 ) can be a treatment for breast cancer. Hence, cytarabine may produce a similar effect to gemcitabine and could be an optional chemotherapeutic drug for treating cervical cancer by targeting rs5030743 and rs1130609 or other similar SNPs.…”

“…Mcl-1 expression in breast cancer cells is down-regulated by BAY 61-3606, making cancer cells more susceptible to TRAIL-mediated apoptosis [ 45 ]. It is feasible for LM patients with breast cancer to respond rapidly to the quick start of intracellular liposomal cytarabine treatment [ 46 ]. BI-2536 is a mitotic inhibitor that blocks the growth and invasion of cancer cells [ 47 ].…”

“…44 It has been recommended that methotrexate, cytarabine, liposomal cytarabine and thiotepa are the most common options for IT therapy. [45][46][47][48][49][50][51][52] Radiotherapy is an alternative treatment targeting bulky, symptomatic disease sites, particularly in the spine, with more efficacy and less toxicity than IT. 53,54 Despite not prolonging patient survival, radiotherapy can help facilitate IT use by restoring CSF flow and relieving hydrocephalus.…”
