The success of biological control of the codling moth, Cydia pomonella, with commercial products based on C. pomonella granulovirus (CpGV) isolates relies both on the ingestion of a sufficient amount of occlusion bodies (OBs) between their hatching from the egg and the penetration inside the fruits, and the correct replication of the virus in the insect larvae. Larvae resistant to one isolate, CpGV-M are frequently found in European orchards. In recent years, efforts have been devoted to obtaining virus isolates able to control resistant insects, among them, CpGV-R5. Ingestion of CpGV-M by a resistant larva does not allow replication of CpGV-M. Ingestion of CpGV-R5 results in virus replication and larval death. Ingestion of a mixture of CpGV-M and CpGV-R5 allows replication of both genotypes and better insect control. Both CpGV-M and CpGV-R5 are present in the commercial product Carpovirusine® Evo 2 (NPP, Arysta LifeSciences). No recent data have been collected concerning the actual amount of virus ingested by the neonate larvae. As mixed infection CpGV-M and CpGV-R5 provide better protection, this is an interesting property to explore to improve the strategy of treatment in orchards. We analysed the distribution of virus droplets on leaves for two orchards using two different dispersion techniques and leaves sprayed in laboratory conditions. We observed the mortality of larvae allowed to feed on these contaminated leaves for selected amounts of time. We then used High resolution melting (HRM) to estimate the occurrence of each genotype alone and mixed genotypes in individual larvae.

4RESUMO -Os baculovírus são vírus patogênicos a insetos, encontrados principalmente na ordem Lepidoptera. A família Baculoviridae é taxonomicamente dividida em dois gêneros: Nucleopolyhedrovirus e Granulovirus, que diferem pela morfologia do corpo de oclusão. Os nucleopoliedrovírus (NPV) possuem corpos de inclusão poliédrica (PIB), contendo múltiplas partículas virais, enquanto os granulovírus (GV) contêm, em geral, partículas únicas, ocluídas em corpos protéicos de forma ovóide. Durante o ciclo de vida são produzidos dois tipos de progênies virais: BV (Budded Virus) e PDV (Polyhedra Derived Virus), que são essenciais para o processo de infecção e propagação do vírus. Os baculovírus têm sido empregados no controle de pragas e, por serem específicos e restritos a invertebrados, são considerados agentes seguros de controle biológico. Recentemente têm sido amplamente utilizados como vetor de expressão de genes heterólogos por produzirem e processarem, em grande quantidade, proteínas de procariotos e eucariotos. Além disso, técnicas de DNA recombinante têm permitido a produção de inseticidas virais geneticamente modificados. Este trabalho constitui uma revisão sobre a taxonomia, estrutura, replicação e biologia molecular de baculovírus e sobre seu uso como bioinseticida no Brasil.

Physical maps and virulence of Anticarsia gemmatalis nucleopolyhedrovirus genomic variants

Seventeen plaque purified isolates of two viral preparations of Anticarsia gemmatalis multiple nucleopolyhedrovirus (AgMNPV), were analyzed in terms of the genomic changes after digestion of their DNAs with HindIII and PstI restriction enzymes. The 1979 AgMNPV wild type preparation (AgMNPV-'79) resulted in six different variants and the 1985 viral commercial preparation (AgMNPV-'85), in eleven. The genomic variation of all the isolates was mapped showing that those from 1985 presented more heterogeneity with changes mapped in additional sites in comparison to the AgMNPV-'79 variants. Their virulence was compared by infecting two Lepidopteran cell lines, Spodoptera frugiperda (IPLB-SF-21AE) and Anticarsia gemmatalis (UFL-AG-286). The results indicated that there was some difference in virulence within the AgMNPV-'85 variants. This commercial preparation had been applied in soybean fields in Brazil over several years to control the velvetbean caterpillar defoliation.
