Efficacy of a Once‐Daily Formulation of Carvedilol for the Treatment of Hypertension

Abstract: 4.4±0.9, 7.9±0.9, and 9.6±0.9 mm Hg, respectively (P≤.001, trend test for all carvedilol CR doses with placebo). Corresponding 24-hour systolic blood pressure changes were 0.6±1. 4, 6.8±1.3, 10.1±1.4, and 12.5±1.3 mm Hg, respectively (P≤.001, trend test for the management of hypertension and for such indications as improving survival and reducing clinical end points in patients with heart failure (HF) or who have experienced an acute myocardial infarction. 1-3 These agents have long been part of antihyperten… Show more

“…ABP treatment differences usually agree well with the result from clinic measures, and treatment was effective when measured by both methods. [26][27][28] The difference between the groups was 8/4 mm Hg for night-time ABP and even higher in the 52 participants with ABP measurements at baseline and during follow-up (online-only Data Supplement). Over 65 years of age, night-time ABP is a better predictor of cardiovascular events than the CBP and other measures of ABP.…”

“…66 Adverse event findings were similar in the placebo and carvedilol CR groups for headache, fatigue, dizziness, and erectile dysfunction. 66,67 The convenience of once-daily dosing combined with a low adverse event profile is a key strategy to improve medication adherence in patients with hypertension and diabetes.…”

“…66 All three doses of carvedilol CR treatment significantly decreased diastolic and systolic blood pressure by study end compared with placebo ( Figure 4). 66 Adverse event findings were similar in the placebo and carvedilol CR groups for headache, fatigue, dizziness, and erectile dysfunction.…”

“…In a recently published study, Weber et al (2006) have shown that once-daily administration of carvedilol CR for 6 weeks in doses ranging from 20 to 80 mg, alone or in combination with other agents, produced sustained diastolic and systolic BP and heart rate reduction compared with placebo and an important BP-lowering effect into the early morning hours. Another study in 122 hypertensive patients, Henderson et al evaluated the side-effect profi le of patients switching from carvedilol twice daily to carvedilol CR.…”
