Efficacy of a novel topical combination of fipronil, (S)-methoprene, eprinomectin and praziquantel against adult and larval stages of Toxocara cati in cats

Abstract: The efficacy of a novel topical combination of fipronil 8.3% (w/v), (S)-methoprene 10% (w/v), eprinomectin 0.4% (w/v), and praziquantel 8.3% (w/v) (BROADLINE(®), Merial) was evaluated against adult and larval Toxocara cati in four controlled studies. All studies included experimentally infected, purpose-bred, short-haired cats. In two studies, 22 or 20 cats harbouring patent infections as confirmed by pre-treatment faecal examination, were included. Within each study, cats were allocated to one of two groups: … Show more

“…Fipronil, a broad-spectrum phenylpyrazole insecticide, has been widely used in pest control and veterinary drugs. 1,2 The bioactivity of fipronil is ascribed to its ability to target γaminobutyric acid (GABA) receptors (GABARs) and act as a noncompetitive blocker of the GABA-gated chloride channels in the central nervous system. 3−5 In addition, fipronil displays greater affinity for insect GABARs than for vertebrate GABARs.…”

“…Both compounds also have appropriate PK parameters (AUC, C max and T max ), that are identical to those of Broadline Ò , a product with proven efficacy against nematodes and cestodes [12,29,35]. This was confirmed by efficacy data obtained with NexGard Ò Combo against both nematodes and cestodes [13] in cats.…”

“…Eprinomectin is a well-known compound used in cattle, sheep and goats [27,33], and cats [12,29]. Praziquantel is a pyrazino-isoquinoline derivative anthelminthic that acts specifically on cestodes and trematodes, and is a well-known compound in veterinary and human medicine, in oral and topical forms [2,3,13,29,32,35,39].…”
