Efficacy of a novel oral chewable tablet containing sarolaner, moxidectin and pyrantel (Simparica Trio™) against natural flea and tick infestations on dogs presented as veterinary patients in Europe

Abstract: Background: A novel chewable oral tablet containing sarolaner, moxidectin and pyrantel (Simparica Trio ™) has recently been developed to provide persistent protection against flea and tick infections for a month, treatment of hookworm and roundworm infections and prevention of heartworm and lungworm disease in dogs. Two field studies were conducted to evaluate the safety and efficacy of Simparica Trio ™ against natural flea and tick infestations on dogs in Europe. Methods: Dogs with natural flea or tick infest… Show more

“…In addition to internal parasites, dogs are also at risk for infestation with fleas and ticks, which can have deleterious effects on their hosts including the transmission of disease agents to both dogs and humans; therefore, regular treatment of dogs at risk for these parasites is recommended [22][23][24][25]. The efficacy of Simparica Trio ™ against flea and tick infestations both under field and laboratory conditions has been demonstrated [26][27][28][29][30].…”

“…The preventative efficacy of Simparica Trio ® for angiostrongylosis has recently been confirmed in placebo-controlled, randomized, masked laboratory studies where dogs were experimentally infected with A. vasorum [37]. Simparica Trio ® , besides preventing angiostrongylosis, also provides effective treatment of flea and/or tick infestations due to the inclusion of sarolaner [38,39] and, due to pyrantel and moxidectin inclusion, efficacy against four gastrointestinal nematode species [40]. Additionally, the 24-48 µg/kg moxidectin dose in the combination is effective in the prevention of heartworm disease caused by Dirofilaria immitis [41].…”

“…In a laboratory study, the number of fleas on dogs orally dosed with afoxolaner was reduced by 100% for at least 22 days [ 48 ]. An oral dose of afoxolaner + milbemycin oxime to naturally infested dogs provided ≥96.1% reductions of fleas at day 30 [ 49 ]. In a field trial with 37 naturally infested dogs in Tampa FL, USA, dogs were provided an oral dose of afoxolaner.…”

“…Both treatments provided >90% anti-attachment of Rhipicephalus sanguineus for at least 14 days [ 48 ]. An oral dose of afoxolaner + milbemycin oxime provided ≥94.4% reductions of ticks on naturally infested dogs at day 30 [ 49 ].…”
