Efficacy and Safety of Two-Drug Regimens with Dolutegravir plus Rilpivirine or Lamivudine in HIV-1 Virologically Suppressed People Living with HIV

Abstract: Background: The high effectiveness and safety of the two-drug (2DRs) strategy using dolutegravir (DTG) plus lamivudine (3TC) have led to international guidelines recommending their use for treatment-naive HIV patients. In virologically suppressed patients, de-escalating from 3DRs to DTG plus either rilpivirine (RPV) or 3TC has shown high rates of virological suppression. Objectives: This study aimed to compare the real-life data of two multicenter Spanish cohorts of PLWHIV treated with DTG plus 3TC (SPADE-3) o… Show more

“…Signi cant e cacy has been achieved in terms of triple treatment, and caused patients into a virological suppressed state according to randomized controlled clinical studies [23,24] TT had caused more toxic side effects, due to the higher number of drug species and complex interactions between drugs [25]. Meanwhile lamivudine-based dual therapies were effective and safe as a switching strategy in patients with an HIV-1 infection who are virologically suppressed[26], with a low virological failure and high viral suppression rates [27]. Comparisons among the interventions in all ranking indicators, only DTG + 3TC displayed signi cantly improved CCFB compared with TT, no signi cant difference was observed in any other comparisons among the interventions.…”
