Efficacy and Safety of COVID-19 Vaccination in Older Adults: A Systematic Review and Meta-Analysis

Abstract: Objective: To analyze the differences in efficacy and safety of different types of novel coronavirus pneumonia (COVID-19) vaccines in different age groups (young adults and elderly). Methods: Randomized controlled trials (RCTs) on COVID-19 vaccine in PubMed, Embase, Web of Science, and Cochrane library were searched by computer, and eight eligible studies were analyzed. Meta-analysis was performed using Stata 16.0 and RevMan5.4 software. Results: The mean geometric titer (GMT) of the virus in the elderly was s… Show more

“…Our findings are consistent with those of several studies suggesting that the elderly may experience fewer adverse events following COVID-19 vaccination than younger people. 37 , 38 Vaccine-related systemic adverse events are related to immune responses, and the incidence of early adverse events is low if the immune response is weak. 39 Lower antibody levels in the elderly after vaccination also confirm this finding.…”

“…Injection site pain, fatigue, and headache are the most reported adverse effects. Rarely, thromboembolic phenomena have been reported with mRNA vaccines and viral vector vaccines 6 . Interestingly, metastatic disease, advanced age, and male sex in patients with solid tumors are independent risk factors that have been shown to reduce antibody response following vaccination.…”

“…Rarely, thromboembolic phenomena have been reported with mRNA vaccines and viral vector vaccines. 6 Interestingly, metastatic disease, advanced age, and male sex in patients with solid tumors are independent risk factors that have been shown to reduce antibody response following vaccination. Vaccine induced lymphadenopathy has been noted in patients with preexisting breast cancer and melanoma, with persistence of lymphadenopathy for more than 6 weeks in 29% of patients.…”

“…This tendency might be related to the difference in antigen presentation and modes of action of both vaccine types [ 104 ]. More importantly, according to a recent systematic review and meta-analysis, giving multiple COVID-19 vaccinations to the elderly is considered effective and safe [ 105 ].…”
